摘 要:目的 探究异绿原酸A对变应性鼻炎(AR)大鼠的炎症抑制作用及机制。方法 36只SD大鼠随机分为对照组(Sham)、模型组(Model)和治疗组(ICOA),每组12只。卵清蛋白致敏制备AR大鼠模型,观察异绿原酸A抗AR的作用,比较各组大鼠行为学评分;HE染色观察大鼠鼻黏膜病理变化;ELISA检测各组大鼠鼻腔灌洗液中EOS%、IFN-γ、IL-4含量及血清OVA-sIgE含量;免疫组化检测鼻黏膜组织中TSLP、IL-4、IFN-γ表达分布情况;Westernblot检测NLRP3、 ASC、 caspase-1蛋白表达量。结果治疗21d起AR症状评分(4.31±0.46)显著低于Model组(6.04±0.70),鼻黏膜病理显著改善,炎症细胞浸润减轻,大鼠体液中OVAsIgE、EOS%及IL-4含量显著降低,IFN-γ表达水平上调,鼻黏膜组织IL-4、TSLP阳性细胞表达量较Model组受到显著抑制,IFN-γ表达量增加,抑制黏膜组织NLRP3、ASC、caspase-1蛋白表达水平。结论 异绿原酸A对AR模型大鼠的治疗作用与调控细胞免疫应答平衡、抑制NLRP3炎性复合体组装相关。Objective To investigate the therapeutic effect of isochlorogenic acid A(ICOA) on inflammation inhibition of allergic rhinitis in rats. Methods 36 SD rats were randomly divided into control group, model group and ICOA group averagely. Oval albumin sensitization was used to establish AR model, the behavioral scores of rats were evaluated, HE staining was used to detect pathological changes. The contents of EOS%, IFN-γ, IL-4 in nasal lavage fluid and OVA-IgE expression level in serum were detected by ELISA. Immunohistochemistry was used to detect the expression and distribution of TSLP, IL-4 and IFN-γ. The expression levels of NLRP3, ASC and caspase-1 proteins were measured by Western blot. Results The symptom score was significantly lower, the pathological change of the nasal mucosa were significantly ameliorated in ICOA group than in model group. ICOA significantly reduced the contents of OVA-sIgE and EOS% and IL-4 in nasal lavage fluid, and upregulated the expression of IFN-γ, inhibited IL-4 and TSLP positive cells in nasal mucosa, and inhibited the expressions of NLRP3, ASC and caspase-1 in the nasal mucosa of rats. Conclusion The therapeutic role of Isochlorogenic acid A is related to regulating cellluar immune response and suppressing NLRP3inflammasome assembly in AR rats.
