摘  要:目的探讨温阳通脉颗粒对糖尿病模型大鼠血管内皮功能的保护作用及作用机制。方法取110只SPF级大鼠,喂养高糖高脂饲料1个月,按体质量腹腔注射链尿佐菌素溶液50 mg/kg,复制糖尿病模型大鼠,将建模成功的大鼠随机分为模型组(等量生理盐水),羟苯磺酸钙组(0.135 g/kg),温阳通脉颗粒高、中、低剂量组(5.000,2.500,1.250 g/kg),各10只;另取10只健康大鼠作为空白组(等量生理盐水)。各组大鼠均灌胃相应药物,每天1次,连续30 d。测定大鼠血糖及血清三酰甘油(TG)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、总胆固醇(TC)、糖化血红蛋白(HbA_(1C))、内皮素(ET)、一氧化氮(NO)的水平;行免疫组化检查,测量大鼠血管内皮生长因子(VEGF)阳性细胞数,并计算阳性细胞面密度和阳性表达率。结果与模型组比较,温阳通脉颗粒高、中、低剂量组大鼠的血糖和血清HbA_(1C),TG,TC,LDL,ET水平及VEGF阳性表达率均显著降低,血清HDL和NO水平均显著升高(P<0.05);各组间阳性细胞面密度比较均无显著差异(P>0.05)。结论温阳通脉颗粒能改善糖尿病模型大鼠的血脂和血糖水平,保护血管内皮功能,其作用机制可能与调节NO和ET的释放平衡、抑制VEGF的表达有关。Objective To investigate the protective effect and mechanism of Wenyang Tongmai Granules on the vascular endothelial function of model rats with diabetes mellitus(DM).Methods A total of 110 SPF rats were selected and fed with high-sugar and high-fat feed for one month.The rats were intraperitoneally injected with 50 mg/kg of streptozotocin(STZ)solution according to their body mass to replicate model rats with DM,then the successfully modeled rats were randomly divided into the model group(equal volume of normal saline),the calcium dobesilate group(0.135 g/kg)and Wenyang Tongmai Granules high-,medium-and low-dose groups(5.000,2.500,1.250 g/kg),with 10 rats in each group.Another 10 healthy rats were taken as the blank group(equal volume of normal saline).The rats in each group were given corresponding drugs by gavage once a day for continuous 30 d.The levels of blood glucose,serum triglyceride(TG),low-density lipoprotein(LDL),high-density lipoprotein(HDL),total cholesterol(TC),glycosylated hemoglobin(HbA_(1C)),endothelin(ET)and nitric oxide(NO)of the rats were determined.The immunohistochemistry examination was used to measure the number of positive cells of vascular endothelial growth factor(VEGF)in rats,and corresponding area density of positive cells and positive expression rate were calculated.Results Compared with those in the model group,the levels of blood glucose,serum HbA_(1C),TG,TC,LDL,ET and the positive expression rates of VEGF in the Wenyang Tongmai Granules high-,medium-and low-dose groups were significantly lower,while the levels of serum HDL and NO in the Wenyang Tongmai Granules high-,medium-and low-dose groups were significantly higher(P<0.05).There was no significant difference in the area density of positive cells in each group(P>0.05).Conclusion Wenyang Tongmai Granules can improve the levels of blood lipid,blood glucose and protect vascular endothelial function of model rats with DM.Its mechanism may be related to the regulation of the release balance of NO,ET and the inhibiton of the expressi
