摘  要:目的探讨局部外用乾坤散对急性软组织损伤模型大鼠血液流变学指标、血栓素A(_(2)TXA_(2))、前列腺素I(_(2)PGI_(2))的影响。方法选取50只SD大鼠采取砸伤法复制急性软组织损伤模型,建模成功后,随机分为模型组,双氯芬酸组(4×10^(-5)g/cm^(2)),乾坤散低、中、高剂量组(0.15,0.30,0.60 g/cm^(2)),各10只;另取10只为空白组。除模型组外,各组大鼠造模2 h后,将对应药物贴敷于患处,每天2次,连续7 d。计算各组大鼠软组织急性损伤评分;苏木精-伊红染色,观察损伤部位肌肉组织病理学形态;判断组织学修复评级;采用全自动血液流变分析仪测定血浆黏度、全血低切黏度、全血高切黏度,并计算红细胞聚集指数;采用酶联免疫吸附法测定TXA_(2)和PGI_(2)水平。结果造模后7 d,与模型组比较,乾坤散低、中、高剂量组大鼠软组织急性损伤评分均显著降低(P<0.05),乾坤散高剂量组大鼠组织学修复评级为优和良的总比例显著升高(P<0.05),乾坤散低、中、高剂量组大鼠全血低切黏度、全血高切黏度及红细胞聚集指数均显著降低(P<0.05);造模后24 h和造模后7 d,乾坤散低、中、高剂量组大鼠急性损伤组织中TXA_(2)/PGI_(2)均接近1.00。结论局部外用乾坤散可改善急性软组织损伤模型大鼠的血液流变学指标,调节TXA_(2)/PGI_(2)平衡,改善损伤组织的微循环。Objective To investigate the effect of the local external application of Qiankun Powder on the hemorheological indexes,thromboxane A_(2)(TXA_(2))and prostaglandin I_(2)(PGI_(2))of model rats with acute soft tissue injury.Methods A total of 50 SD rats were selected to replicate the acute soft tissue injury model by smashing.After successful modeling,the above 50 rats were randomly divided into the model group,the diclofenac group(4×10^(-5 )g/cm^(2)),the Qiankun Powder low-,medium-and high-dose groups(0.15,0.30,0.60 g/cm^(2)),with 10 rats in each group.The other 10 rats were taken as the blank group.Except for the model group,the rats in the each group were applied with the corresponding drugs on the injured part after 2 h of modeling,twice a day for continuous 7 d.The score of acute soft tissue injury of rats in each group was calculated.The hematoxylin-eosin(HE)staining was used to observe the pathological morphology of muscle tissue at the injured part.The histological repair rating was determined.The full-automatic hemorheological analyzer was used to measure the plasma viscosity,the whole blood low-shear viscosity and the whole blood high-shear viscosity,and the erythrocyte aggregation index was calculated.The enzyme-linked immunosorbent assay(ELISA)was used to measure the levels of TXA_(2) and PGI_(2).Results Compared with those in the model group,the scores of acute soft tissue injury in the Qiankun Powder low-,medium-and high-dose groups significantly decreased after 7 d of modeling(P<0.05),while the excellent and good rate of histological repair rating in the Qiankun Powder high-dose group increased significantly after 7 d of modeling(P<0.05).Compared with those in the model group,the whole blood low-shear viscosity,whole blood high-shear viscosity and erythrocyte aggregation index in the Qiankun Powder low-,medium-and high-dose groups significantly decreased after 7 d of modeling(P<0.05).The TXA_(2)/PGI_(2) of the acute injured tissues of rats in the Qiankun Powder low-,medium-and high-dose groups was cl
