摘 要:目的 研究红花Carthamus tinctorius L.的化学成分及其抑制子宫平滑肌收缩作用。方法 采用D101大孔吸附树脂、硅胶、Sephadex LH-20柱色谱及半制备高效液相色谱等方法进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。通过缩宫素致大鼠离体子宫平滑肌痉挛模型考察化合物对子宫平滑肌收缩活动的影响。结果 从中分离得到8个化合物,分别鉴定为N^(1),N^(5),N^(10)-3-对-香豆酰亚精胺(1)、4-乙氧基桂皮酸(2)、顺式对羟基肉桂酸乙酯(3)、反式对羟基肉桂酸乙酯(4)、(-)-methyl dihydrophaseate(5)、plucheol A(6)、N-顺式-阿魏酰酪胺(7)、桃金娘苷(8)。结论 化合物2~4、6、8均为首次从红花属中分离得到。化合物1、7在10、50μmol/L浓度下对离体子宫平滑肌收缩活动有抑制作用。AIM To study the chemical constituents from Carthamus tinctorius L.and their inhibitory effects on uterine smooth muscle contraction.METHODS The extract from C.tinctorius was isolated and purified by column chromatography over D-101 macroporous adsorption resin,silica gel,and Sephadex LH-20,and semi-preparative high performance liquid chromatography,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The rat uterine smooth muscle model induced by oxytocin was used to evaluate the effects of the compounds on the contractile activity of uterine smooth muscle.RESULTS Eight compounds were isolated and identified as N^(1),N^(5),N^(10)-tri-p-coumaroylspermidine(1),4-ethoxycinnamic acid(2),cis-p-hydroxyl ethyl cinnamate(3),trans-p-hydroxyl ethyl cinnamate(4),(-)-methyl dihydrophaseate(5),plucheol A(6),N-cis-feruloyltryptamine(7),and myrtenyl-O-β-D-glucopyranoside(8).CONCLUSION Compounds 2-4,6,and 8 were isolated from the genus Carthamus for the first time.Compounds 1 and 7 have inhibitory effects on the contractile activity of uterine smooth muscle at the concentrations of 10,50μmol/L.
