摘 要:目的:研究银杏叶提取物对小鼠气管的舒张作用,探讨引起其舒张的作用机制。方法:BALB/c鼠处死后分离气管环,将气管环置于恒温通氧灌流系统中,经高钙液平衡后,加入激动剂乙酰胆碱(Ach)100μmol/L收缩气管环,待气管环张力达到平台期后,加入银杏叶提取物,使其浓度梯度依次为10^(-2)、10^(-1.5)、10^(-1)、10^(-0.5)、10^(0)、10^(0.5)、10^(0.75)、10^(1)g/L,将引起气管环最大收缩的最低银杏叶提取物浓度5.62mg/mL作为本次实验的最佳药物浓度并确定剂量曲线。加入100μmol/L Ach使气管环预收缩,待气管环张力达到稳定后,加入L型钙离子通道抑制剂硝苯地平(Nifedipine)、非选择性阳离子通道抑制剂吡唑盐(Pyr3),待气管环达到平台期时加入5.62mg/mL的银杏叶提取物,观察并记录气管环肌张力的变化曲线。另加入80μmol/L的高K^(+)溶液收缩气管环,待气管环张力稳定后,加入银杏叶提取物,观察并记录气管环张力变化。结果:银杏叶提取物对Ach引起的气管平滑肌预收缩有较强的舒张作用,并呈现明显的浓度依赖性。这种强烈的舒张作用主要是银杏叶提取物通过抑制非选择性阳离子通道和L型钙离子通道介导的钙离子内流实现。结论:银杏叶提取物具有舒张气管的功效,具备作为支气管扩张剂的潜能。Objective:To investigate the diastolic effect of Ginkgo biloba extract(abbreviated EGB)on tracheal ring in mice and explore its mechanism.Methods:BALB/c mice were experiment,the lowest EGB concentration of 5.62mg/mL,which caused the maximum contraction of tracheal ring,was taken as the optimal drug concentration in this experiment.100μmol/L Ach was added into the irrigation tank to preshrink the tracheal ring.Then nifedipine was added as L-type calcium channel inhibitor and Pry3as non-selective cationic channel inhibitor.When the tracheal ring reached the plateau stage,5.62mg/mL of EGB was added to observe and record the change curve of tracheal ring tension and then the tracheal ring was contracted with 80μmol/L high potassium solution.EGB was added to observe and record the changes of tracheal ring tension after the tracheal ring tension was stabilized.Results:EGB had a strong diastolic effect on the precontraction of tracheal smooth muscle induced by Ach,and the effection was concentration dependent.This strong diastolic effect was mainly achieved by inhibiting calcium ion influx of the non-selective cationic channel and L-type calcium channel.Conclusion:EGB can dilate trachea and have the potential as a bronchodilator.
