摘  要:目的:观察不同纯度枸杞多糖(LBP)对光诱导人视网膜色素上皮(RPE)细胞损伤的抗氧化作用。方法:体外培养人视网膜色素上皮细胞(ARPE-19),建立光诱导下hRPE细胞损伤模型,加入不同纯度的枸杞多糖(50%、65%)对hRPE细胞进行干预并于24h后进行相应指标检测。CCK-8法检测细胞活力;流式细胞术(FCM)检测细胞内活性氧自由基(ROS)、细胞超氧化物歧化酶(SOD)、丙二醛(MDA)的含量。实验组分为正常组、模型组、不同纯度枸杞多糖(50%、65%)干预组。结果:与正常组比较,模型组细胞存活率明显降低,SOD活力明显下降,ROS、MDA明显升高(P<0.01);与模型组比较,不同纯度枸杞多糖(50%、65%)干预组细胞存活率明显升高,SOD活力明显上升,ROS、MDA呈下降趋势(P<0.01);65%枸杞多糖组较50%枸杞多糖组细胞存活率明显升高,SOD活力呈上升趋势,ROS、MDA明显下降(P<0.01)。结论:枸杞多糖可保护光诱导的hRPE细胞氧化损伤,随着枸杞多糖纯度提高,其保护作用更显著。Objective:To observe the antioxidant effects of Lycium barbarum polysaccharides(LBP)of different purity on light-induced damage of human retinal pigment epithelium(RPE)cells.Methods:Human retinal pigment epithelial cells(ARPE-19)were cultured in vitro to establish hRPE cell light damage model.The hRPE cells were intervened with different purity Lycium barbarum polysaccharides(50%,65%),and the corresponding indicators were detected after 24hours of light.CCK-8cell viability test;Flow cytometry(FCM)was used to detect the content of reactive oxygen free radicals(ROS)in cells;the content of superoxide dismutase(SOD)and malondialdehyde(MDA)in cells was detected.The experimental components were normal group,model group,and intervention group of Lycium barbarum polysaccharides(50%,65%)with different purity.Results:Compared with the normal group,the cell survival rate of the model group was significantly reduced,SOD activity was significantly decreased,and ROS and MDA were significantly increased(P<0.01);compared with the model group,the cell survival rate of the Lycium barbarumpolysaccharides(50%,65%)intervention group was significant increased,SOD activity increased significantly,ROS and MDA showed a downward trend(P<0.01);compared with 50% Lycium barbarum polysaccharides group,cell survival rate of 65% Lycium barbarum polysaccharides group increased significantly,SOD activity showed an upward trend,and ROS and MDA significantly decline(P<0.01).Conclusion:Lycium barbarum polysaccharides can protect hRPE cells from light-induced oxidative damage,and its protective effect becomes more significant as the purity of Lycium barbarum polysaccharides increases.
