摘  要:本试验旨在通过诱变选育得到一株高产二十二碳六烯酸(DHA)的裂殖壶藻藻株,为进一步开发缓解幼龄动物腹泻的新型饲料奠定基础。使用裂殖壶藻原始藻株ATCC20888,通过常压室温等离子体对藻株进行诱变,再进一步通过建立丙二酸、2,2’-联吡啶及丙二酸-2,2’-联吡啶复合筛选方法对诱变藻株进行快速筛选,对筛选突变藻株进行摇瓶发酵,利用磷酸香草醛法和高效气相色谱法对突变藻株的油脂、DHA产量快速检测,得到高产DHA的藻株。结果表明:丙二酸、2,2’-联吡啶及丙二酸-2,2’-联吡啶复合筛选均可以提高裂殖壶藻藻株的平均DHA产量,较原始藻株分别提高了32.20%、25.01%和29.71%。通过丙二酸-2,2’-联吡啶复合筛选得到的诱变藻株I-F-9其DHA含量(54.56%)及产量(10.88 g/L)分别比原始菌株增加了74.62%和61.17%(P<0.05)。由此可见,通过常压室温等离子体诱变选育的裂殖壶藻藻株I-F-9较原始藻株DHA含量及产量均有显著提高,且其DHA产量处于目前同类研究前列。This study aimed to obtain a high docosahexaenoic acid(DHA)-production Schizochytrium sp.strain through mutation breeding,to provide the basis for developing dietary antibiotic alternatives to alleviate diarrhea occurred in calves,especially the calves before weaning.Schizochytrium sp.ATCC20888 as an original strain,which was mutagenized by atmospheric and room-temperature plasma.Mutagenized strains were selected by malonic acid,2,2’-bipyridine and malonic acid-2,2’-bipyridine combination rapid screening protocols.The selected mutant strains were fermented in shake flasks,and then were evaluated for total oil and DHA production through vanillin phosphate detection and high-performance gas chromatography,to obtain high DHA production strains.The results showed that the average DHA production of Schizochytrium sp.strains screened by malonic acid,2,2’-bipyridine and malonic acid-2,2’-bipyridine combination were all improved,which was improved by 32.20%,25.01%and 29.71%compared with the original strain.The DHA content(54.56%)and production(10.88 g/L)of mutant strain I-F-9 screened by malonic acid-2,2’-bipyridine combination were improved by 74.62%and 61.17%compared with the original strain(P<0.05).In conclusion,the DHA content and production of Schizochytrium sp.strain I-F-9 mutagenized by atmospheric and room temperature plasma are significantly higher than the original strain,and the DHA production is relatively higher compared with other similar studies.
