摘 要:为延长朱顶红离体保存时间、提高存活率,研究了甘露醇、蔗糖、矮壮素、脱落酸对朱顶红试管苗离体保存及恢复生长的影响。结果表明:在培养室温度(15±2)℃、光照强度2500~3000 lx、光照时间10 h/d的条件下,MS培养基中添加20~40 g/L甘露醇能有效抑制朱顶红植株生长,保存5个月后恢复培养2个月的死亡率为0%~40%;75 g/L蔗糖能抑制朱顶红生长发育,离体保存5个月、恢复生长2个月后存活率达100%;0.5~4.0 mg/L ABA能明显抑制朱顶红植株生长,但恢复生长培养后死亡率较其他几种添加物高,恢复生长培养2个月后死亡率最高达50%。10~50 mg/L矮壮素的添加对朱顶红生长抑制效果较差。In order to prolong the in vitro preservation time and improve the survival rate,the effects of mannitol,sucrose,chlormequat chloride and abscisic acid on the in vitro preservation and recovery growth of Hippeastrum tube seedlings were studied.The results showed that the addition of 20~40 g/L mannitol could effectively inhibit the growth of Hippeastrum at(15±2)℃ under the 10 h photoperiod culture condition of 2500~3000 lx,and the mortality rate was 0%~40% after 6 months in vitro storage and 2 months recovery growth.The 75 g/L sucrose could inhibit the growth and development of Hippeastrum,and the survival rate was 100% after 5 months in vitro storage and 2 months recovery growth.The 0.5~4 mg/L ABA could significantly inhibit the growth of Hippeastrum,but the mortality rate was higher than other osmotic regulatory substances.The mortality rate was up to 50% after 5 months in vitro storage and 2 months recovery growth.The 10~50 mg/L chlormequat chloride had a poor inhibition effect on plant growth.
