摘 要:肠道病毒A组71型(Enterovirus 71,EV⁃A71)是引起婴幼儿手足口病(Hand,foot and mouth disease,HFMD)的主要病原体,研究表明EV⁃A71感染诱发的中枢神经系统(Central nervous system,CNS)并发症与EV⁃A71患儿机体的免疫失调有着密切的关系。白细胞介素⁃2(interleukin⁃2,IL⁃2)是一种能激活T细胞,并维持T细胞分化和增殖的T细胞生长因子,同时也参与炎症或自身免疫性反应。本研究旨在研究低剂量IL⁃2(500 IU/只)对EV⁃A71感染乳鼠模型的影响。研究结果发现,注射低剂量IL⁃2能显著改善EV⁃A71感染BALB/c乳鼠的存活率和疾病指数,能明显减轻EV⁃A71感染BALB/c乳鼠的脑部和肺部的病理损伤,能显著降低EV⁃A71感染Balb/c乳鼠脑部的病毒载量,并且能显著下调IL⁃1β、IL⁃6、IL⁃18和TNF⁃α的表达。本研究的结果初步提示了低剂量IL⁃2用于治疗HFMD的有效性,为IL⁃2用于EV⁃A71感染导致的HMFD患者的临床治疗提供了理论依据。Enterovirus 71(EV⁃A71)is the main pathogen causing hand,foot and mouth disease(HFMD)in infants and young children.Studies have shown that EV⁃A71 infection⁃induced central nervous system(CNS)complications are closely related to immune disorders in children with EV⁃A71.Interleukin(IL)⁃2 is a T⁃cell growth factor that can activate and maintain the differentiation and proliferation of T cells.IL⁃2 is also involved in inflammatory or autoimmune responses.We investigated the effect of low⁃dose(500 IU/mouse)IL⁃2 on an EV⁃A71⁃infected model in suckling mice.Injection of low⁃dose IL⁃2 could lengthen survival and improve the disease index significantly,as well as significantly reduce pathological damage to the brain and lungs of EV⁃A71⁃infected BALB/c suckling mice.Low⁃dose IL⁃2 could also significantly reduce the viral load in the brain and significantly downregulate expression of IL⁃1β,IL⁃6,IL⁃18 and tumor necrosis factor⁃αin EV⁃A71⁃infected BALB/c suckling mice.Our preliminarily study suggests the efficacy of low⁃dose IL⁃2 in HFMD treatment,and provides a theoretical basis for use of IL⁃2 to treat HMFD caused by EV⁃A71 infection.
