摘  要:目的通过对人参属5种不同药性的中药,即三七、红参、人参、西洋参及人参叶中的人参皂苷含量与药性进行相关性分析,并通过Na^(+)/K^(+)-ATPase活力测定进行验证,建立人参单体皂苷的药性(气)评价方法。方法采用双变量相关分析人参单体皂苷与中药药性相关性,灰色关联法评价其对人参属中药药性(气)的贡献,运用网络药理学预测代谢通路,测定对人肝细胞LO2与猪大脑皮层Na^(+)/K^(+)-ATPase活力影响评价人参单体皂苷药性。结果双变量相关分析结果表明,与温热和寒凉药性相关皂苷的相关程度分别为Rf>R1>Rg3>Rg2>Rb1>Ro和Rb2>Re>Rd>F11。灰色关联度分析结果表明,其贡献程度顺序为F11>Re>Rg2>Rd>Rb2>Rb1>Rg1>Rc>Rg3>R1>Rf>Ro。对Na^(+)/K^(+)-ATPase活力影响表明,人参皂苷Rb1、Rb3、Rc、Rf、Rg2、Rg1、Rh1、Rg3、R1与Ro为温热药性,能增强Na^(+)/K^(+)-ATPase活力,人参皂苷Rb2、Rd、Rh2、Re及齐墩果酸为寒凉药性,能抑制Na^(+)/K^(+)-ATPase活力。结论利用Na^(+)/K^(+)-ATPase活力结合人参皂苷含量与中药药性相关性分析进行人参单体皂苷的药性(气)归属,方法有一定可行性。Aim To establish the evaluation methods of the medicine nature of ginsenosides by correlation analysis between medicine nature and ginsenoside contents in five Panax herbs with different medicine nature and the measurement of their effects on Na^(+)/K^(+)-ATPase activities.Methods The contents of ginsen-osides in Sanqi,red ginseng,ginseng,American ginseng and ginseng leaves in the existing literature were collected,and the medicinal nature was assigned by vector methods.The medicine nature of ginsenosides and the contribution of ginsenoside to the medicine nature were evaluated through bivariate correlation analysis and grey correlation degree respectively.The effects of ginsenosides on the Na^(+)/K^(+)-ATPase activities of LO2 cells and in pig cerebral cortex were measured to evaluate the medicine nature of ginsenosides.Results Correlation results indicated that the order of correlation coefficients of ginsenosides to the warm and hot medicine nature was Rf>R1>Rg3>Rg2>Rb1>Ro,while the order of correlation coefficients of ginsenosides to the cool and cold medicine nature was Rb2>Re>Rd>F11.Grey correlation degree analysis showed that the contribution of ginsenoside to the medicine nature was F11>Re>Rg2>Rd>Rb2>Rb1>Rg1>Rc>Rg3>R1>Rf>Ro.The effects of ginsenosides on Na^(+)/K^(+)-ATPase activities showed that the substance basis of the warm medicine nature was ginsenoside Rb1,Rb3,Rc,Rf,Rg2,Rg1,Rh1,Rg3,R1 and Ro,which increased the activity of Na^(+)/K^(+)-ATPase.While the cold and cool medicine nature were ginsenoside Rb2,Rd,Rh2,Re and oleanolic acid,which inhibited the activity of Na^(+)/K^(+)-ATPase.Conclusion It is feasible to evaluate the medicine nature of ginsenoside according to Na^(+)/K^(+)-ATPase activities and correlation analysis between medicine natures and ginsenosides contents.
