摘 要:目的观察款冬花散对支气管扩张症痰热蕴肺证模型大鼠气道黏液高分泌的影响。方法70只大鼠随机分为空白组,款冬花散自身对照组,模型组,罗红霉素组,款冬花散低、中、高剂量干预组,每组10只。予绿脓杆菌液气管注入制备支气管扩张症大鼠模型。采用款冬花散干预2周后取材,AB-PAS染色观察大鼠肺组织气道黏液分泌情况、肺泡灌洗液(BALF)中NE、Muc5ac水平及肺组织NE、Muc5ac蛋白和mRNA表达水平。结果与空白组相比,模型组大鼠气道杯状细胞增多同时有大量黏液形成,BALF的NE、Muc5ac水平及肺组织NE、Muc5ac蛋白和mRNA表达水平明显增加(P<0.01)。款冬花散低、中、高剂量组杯状细胞数均有所降低,且气道黏液分泌减少;款冬花散减轻大鼠BALF的NE、Muc5ac水平,下调肺组织NE、Muc5ac蛋白和mRNA表达水平,且以高剂量组较为明显(P<0.05或P<0.01)。结论款冬花散在一定程度上降低支气管扩张症痰热蕴肺证模型大鼠气道黏液高分泌状态,并呈现一定的量效关系。Objective To observe the effect of Kuandonghua San(款冬花散)on airway mucin hypersecretion in rats for bronchiectasis with the phlegm-heat stagnation syndrome.Methods A total of 70 rats were randomly divided into the blank group,the self-control group,the model group,the roxithromycin group,the Kuandonghua San treatment group at low,middle and high dosage,10 in each group.The rat model was induced by intratracheal injection of Pseudomonas aeruginosa dilution.Rats in the treatment group were treated with Kuandonghua San for 2 weeks.AB-PAS staining of lung tissue,concentrations in bronchoalveolar lavage fluid(BALF)and expressions of protein and mRNA in lung tissue including NE and Muc5ac were evaluated.Results Compared with the blank group,goblet cell proliferation,concentrations in BALF,and expressions of protein and mRNA in lung tissue including NE and Muc5ac of the model group were significantly increased(P<0.01).Compared with the model group,goblet cell proliferation,concentrations in BALF and expressions of protein and mRNA in lung tissue with regard to NE and Muc5ac of the Kuandonghua San treatment group were significantly decreased,which the effect of high dosage(P<0.05 or P<0.01)was superior to low and middle dosage.Conclusion Kuandonghua San can reduce airway mucin hypersecretion in rats for bronchiectasis with the phlegm-heat stagnation syndrome,and it presents a certain quantity-effect relationship.
