摘 要:目的探讨生品和蜜炙桑白皮分别作用于经方泻白散祛痰平喘的作用及机制。方法将昆明小鼠随机分为模型对照组、盐酸氨溴索组、生品与蜜炙桑白皮的泻白散0.57、1.14、2.28、4.55 g·kg^(-1)剂量组。连续10 d灌胃给药,采用气管酚红排泌实验检测小鼠酚红排泌量;Wistar大鼠随机分为空白对照组、模型对照组、地塞米松组、生品和蜜炙桑白皮的泻白散3.15、9.45 g·kg^(-1)剂量组,造模大鼠于第1、7天腹腔注射卵蛋白混悬液1 mL致敏,从第15天起,每天激发造模,共5 d,第20天记录大鼠10 min内出现Ⅱ级哮喘的潜伏期,检测肺泡灌洗液(BALF)中嗜酸粒细胞数(EOS)、血清中一氧化氮(NO)含量以及肺组织病理学变化。结果与模型对照组相比,生品桑白皮泻白散1.14、2.28g·kg^(-1)剂量组以及蜜炙桑白皮的泻白散各剂量组均可显著升高小鼠气管酚红排泌量。与空白对照组相比,模型对照组大鼠引喘潜伏期明显缩短,肺泡灌洗液的EOS、血清中NO的含量均显著增加,哮喘症状典型。支气管腔狭窄,黏膜上皮中杯状细胞增多,并有嗜酸性粒细胞浸润,气道黏膜下组织水肿,支气管内分泌物潴留,肺泡间实变区较正常肺泡间隔面积明显扩大。生品与蜜炙桑白皮的泻白散9.45 g·kg^(-1)剂量组均能显著延长大鼠哮喘潜伏期,减少BALF中EOS,降低血清中NO的含量,明显改善大鼠肺组织病理学变化。与相同剂量生品组比较,蜜炙桑白皮的泻白散剂量组大鼠哮喘潜伏期增长,EOS含量减少。结论生品与蜜炙桑白皮的泻白散剂量组均能明显降低哮喘大鼠BALF中炎性细胞数量,降低血清中NO含量,减轻气道炎症,增强祛痰能力,改善哮喘症状,且蜜炙桑白皮的泻白散剂量组较生品的效果更好。Objective To explore the effect and mechanism of raw and honey fried Sangbaipi(Mori Cortex)on expectorant and antiasthmatic effect of Xiebai Powder(泻白散).Methods KM mice were randomly divided into model control group,ambroxol hydrochloride group,raw and honey fried Sangbaipi(Mori Cortex)Xiebai Powder 0.57,1.14,2.28,4.55 g·kg^(-1) dose groups.Wistar rats were randomly divided into blank control group,model control group,dexamethasone group,raw product and honey fried Sangbaipi(Mori Cortex)Xiebai Powder 3.15 g·kg^(-1) and 9.45g·kg^(-1) dose groups.The model rats were intraperitoneally injected with ovalbumin suspension for 1 hour on the first and seventh days.On the 20th day,the latent period of gradeⅡasthma within 10 minutes was recorded,eosinophil count(EOS)in bronchoalveolar lavage fluid(BALF),serum nitric oxide(NO)content and lung histopathological changes were detected.Results Compared with the model control group,raw Sangbaipi(Mori Cortex)Xiebai Powder(1.14 g·kg^(-1),2.28 g·kg^(-1))and honey fried Sangbaipi(Mori Cortex)Xiebai Powder(all dose groups)could significantly increase the phenol red excretion in trachea of mice.Compared with the blank control group,the latent period of asthma induction in the model control group was significantly shortened,the content of EOS in BALF and NO in serum were significantly increased,and the symptoms of asthma were typical.There were stenosis of bronchi,increase of goblet cells in mucosal epithelium,thickening of basal layer,hypertrophy of smooth muscle,infiltration of eosinophils,edema of airway submucosa,retention of secretion in bronchus,and enlargement of consolidation area between alveoli.Both raw and honey fried Sangbaipi(Mori Cortex)Xiebai Powder(9.45 g·kg^(-1))could significantly prolong the incubation period of asthma,reduce EOS in BALF,reduce the content of NO in serum,and significantly improve the pathological changes of lung tissue in rats.Compared with the same dose of raw material group,the asthma incubation period and EOS of Xiebai Powder group r
