摘  要:目的选取“百会”“印堂”及双侧“足三里”穴进行电针干预,探讨电针对大脑中动脉栓塞(MCAO)模型大鼠脑缺血组织内VGLUT1表达的影响及对CaM/CaMKⅡ信号通路的影响。方法线栓法制备MCAO模型大鼠,依据神经功能评分标准,造模成功后的30只大鼠按照随机数字表法分为假手术组、模型组和电针组,每组10只。造模成功后2 h,选取“百会”“印堂”及双侧“足三里”穴对电针组进行治疗,连续电针7 d。通过Morris水迷宫实验完成对大鼠学习和记忆能力的检测;HE染色法观察各组大鼠脑组织的形态学改变;免疫组化法检测各组大鼠脑缺血组织内VGLUT1表达水平;ELISA法检测各组大鼠脑缺血组织内CaM、CaMKⅡ表达水平。结果经电针治疗后,电针组大鼠神经功能评分较模型组显著降低(P<0.05);Morris水迷宫实验:与模型组相比,电针组大鼠穿过逃生平台的次数明显增多(P<0.01),其逃避潜伏期明显缩短(P<0.05);HE染色:模型组和电针组脑组织细胞均有不同程度的坏死或损伤,但电针组的组织形态学有明显改善;免疫组化法:与模型组比较,电针组大鼠脑缺血组织内VGLUT1表达水平明显升高(P<0.05)。ELISA法:与假手术组比较,模型组和电针组大鼠脑缺血组织内CaM、CaMKⅡ表达水平显著升高(P<0.05)。与模型组比较,电针组大鼠脑缺血组织内CaM、CaMKⅡ表达水平明显降低(P<0.05)。结论电针“百会”“印堂”及双侧“足三里”穴能够明显改善脑缺血再灌注大鼠的学习和记忆能力,上调VGLUT1表达水平,增强突触传递效能,且能够抑制CaM/CaMKⅡ信号通路的活化,对脑组织产生一定的保护作用,对脑缺血再灌注损伤在临床上的治疗科学性提供了实验依据。Objective To select Baihui(DU20),Yintang(EX-HN3)and Zusanli(ST36)on both sides for electroacupuncture intervention,and to explore the effect of electroacupuncture on VGLUT1 expression in cerebral ischemia tissue of middle cerebral artery embolism(MCAO)model rats and its effect on CaM/CaMKⅡsignal pathway.Methods MCAO model rats were made by suture method,and 30 rats were divided into sham operation group,model group and electroacupuncture group with 10 rats in each group.Two hours after successful modeling,the electroacupuncture group was treated with Baihui(DU20),Yintang(EX-HN3)and Zusanli(ST36)on both sides for 7 days.The learning and memory ability of rats was tested by Morris water maze test The morphological changes of brain tissue in each group were observed by HE staining.Immunohistochemical method was used to detect the expression level of VGLUT1 in cerebral ischemia tissues of rats in each group.The expression levels of CaM and CaMKⅡin cerebral ischemia tissues of rats in each group were detected by ELISA.Results After electroacupuncture treatment,the neurological function score of rats in electroacupuncture group was significantly lower than that in model group(P<0.05).Morris water maze test:Compared with the model group,the times of rats crossing the escape platform in electroacupuncture group were significantly increased(P<0.01),and the escape latency was significantly shortened(P<0.05).HE staining:the brain cells in model group and electroacupuncture group were necrotic or damaged to varying degrees,but the histomorphology of electroacupuncture group was improved obviously;Immunohistochemical method:compared with the model group,the expression level of VGLUT1 in cerebral ischemia tissue of rats in electroacupuncture group was significantly higher(P<0.05).ELISA method:Compared with sham operation group,the expression levels of CaM and CaMKⅡin cerebral ischemia tissues of rats in model group and electroacupuncture group were significantly higher(P<0.05).Compared with the model group,the expression
