摘 要:目的探究“原络通经”针法防治痴呆小鼠认知功能障碍的作用机制。方法SAMP8小鼠18只随机分成3组:对照组、“原络通经”针刺组、百会针刺组,每组6只;以SAMR1同品系正常小鼠6只作为对照,通过Morris水迷宫检测治疗后小鼠空间学习记忆能力的变化,TUNEL检测小鼠脑海马组织的细胞凋亡,ELISA检测各组小鼠海马组织中Aβ40、sAPPα、sAPPβ表达,探究“原络通经”针法改善痴呆小鼠空间学习记忆能力的作用机制。结果(1)与SAMR1小鼠比较,SAMP8小鼠出现明显的学习记忆功能障碍;与SAMP8空白组比较,“原络通经”针刺组和百会针刺组小鼠逃避潜伏期均显著缩短(P<0.01);“原络通经”针刺组与百会针刺组比较,小鼠在治疗后水迷宫检测逃避潜伏期明显缩短,且穿过原平台所在位置的次数明显增多(P<0.01);与百会针刺组比较,“原络通经”针刺组小鼠逃避潜伏期缩短,且穿过原平台所在位置的次数增加(P<0.01)。(2)SAMR1组小鼠海马组织细胞形态饱满,形态正常,虽有细胞凋亡,但数量少,SAMP8空白组小鼠海马组织凋亡数量明显增多,与SAMR1比较差异具有统计学意义(P<0.01);与SAMP8空白组比较,“原络通经”针刺组和百会针刺组小鼠海马组织细胞凋亡数量明显减少;与百会针刺组比较,“原络通经”针刺组小鼠海马组织细胞凋亡数量显著减少,差异具有统计学意义(P<0.01)。(3)与SAMR1组比较,SAMP8组小鼠海马组织中Aβ40蛋白表达增加,而sAPPα、sAPPβ表达降低,差异具有统计学意义(P<0.01);“原络通经”针刺组和百会针刺组小鼠海马组织中Aβ40表达明显降低,而sAPPα、sAPPβ表达增加;与百会针刺组比较,“原络通经”针刺组小鼠海马组织中Aβ40含量降低,而sAPPα、sAPPβ表达增加,差异具有统计学意义(P<0.01)。结论“原络通经”针刺法与百会针刺法治疗均可以改善痴呆小鼠的学习记忆能力,“原络通经”针刺法疗效Objective To explore the mechanism of“Yuanluo Tongjing”acupuncture to prevent cognitive dysfunction in mice with dementia.Methods Eighteen SAMP8 mice were randomly divided into three groups:control group,“Yuanluo Tongjing”acupuncture group,and Baihui acupuncture group,with 6 mice in each group;Six SAMR1 normal mice were used as the control group.The changes of spatial learning and memory ability of mice after treatment were detected by Morris water maze.Cell apoptosis in hippocampal tissues of mice was detected by TUNEL.The expressions of Aβ40,SAPPαand SAPPβin the hippocampus of mice in each group were detected by ELISA.To explore the mechanism of“Yuanluo Tongjing”acupuncture to improve the spatial learning and memory ability of dementia mice.Results(1)Compared with SAMR1 mice,SAMP8 mice showed significant learning and memory dysfunction;Compared with SAMP8 blank group,the escape latency of mice in“Yuanluo Tongjing”acupuncture group and Baihui acupuncture group was significantly shortened(P<0.01);Compared with Baihui acupuncture group,“Yuanluo Tongjing”acupuncture group significantly shortened the escape latency of water maze detection,and increased the number of times of crossing the original platform(P<0.01).Compared with Baihui acupuncture group,the escape latency of mice in“Yuanluo Tongjing”acupuncture group was shortened,and the number of times of crossing the original platform was increased(P<0.01).(2)The morphology of hippocampal cells in SAMR1 group was full and normal.Although there was apoptosis,the number of hippocampal cells in SAMR1 group was less.Compared with SAMR1 group,the number of hippocampal cells in SAMP8 group was significantly increased,and the difference was statistically significant(P<0.01);Compared with SAMP8 group,the number of hippocampal cell apoptosis in“Yuanluo Tongjing”acupuncture group and Baihui acupuncture group was significantly reduced;Compared with Baihui acupuncture group,the number of apoptosis in the hippocampal tissue of mice was significant
