摘 要:目的探究人肠道菌群相关代谢物丁酸(butyric acid,BA)对人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,hUC-MSCs)的细胞因子分泌的影响。方法用含不同浓度丁酸(0.02、0.08、0.32、1.25和5.00 mmol/L)的培养基培养细胞24、48和72 h,采用ELISA法检测上清液中白细胞介素6(interleukin 6,IL-6)表达情况,以不含丁酸的完全培养基作为对照组。结果培养24 h,中等浓度区IL-6表达增加,以浓度为1.25 mmol/L时增加最明显,低、高浓度组与对照组比较差异无统计学意义;培养48 h,低浓度组IL-6表达下降,中间浓度区的1.25 mmol/L浓度组及高浓度组表达量增加;培养72 h,低浓度区IL-6表达量下降且差异有统计学意义,中间浓度区表达增加,高浓度组呈下降趋势但差异无统计学意义。结论丁酸对hUC-MSCs的IL-6表达有量效性和时效性,因此丁酸的不同量效和时效对hUC-MSCs的生理性和病理性功能调节可能会有不同效应。Objective To observe the effect of human intestinal microflora-related metabolite butyric acid(BA)on the cytokine secretion of human umbilical cord mesenchymal stem cells(hUC-MSCs).Methods The hUC-MSCs were cultured in stem cell culture medium containing different concentrations of butyric acid(0.02 mmol/L,0.08 mmol/L,0.32 mmol/L,1.25 mmol/L,5.00 mmol/L)for 24,48 and 72 hours respectively to detect the expression of IL-6 in the culture medium by using ELISA,and the complete medium without BA was used as the control group.All the observation groups were compared with the control group.Results At 24 hours,the expression of IL-6 in the moderate concentrations increased,and the increase was the most obvious when the concentration was 1.25 mmol/L.The low and high concentration groups were not statistically significant compared with the control group.At 48 hours,the expression of IL-6 in the low concentration group decreased,and that in the moderate concentrations including 1.25 mmol/L and the higher concentration group increased.All the concentration groups were statistically different compared with the control group.At 72 hours,the expression level of IL-6 in the low concentrations decreased and were statistically different compared with the control group.The expression level of IL-6 in the moderate concentrations increased,and that in the high concentration group showed a downward trend.Two concentration groups were not statistically different compared with the control group.Conclusion The expression of IL-6 in hUC-MSCs is related to both dose-and time-dependent effects of butyric acid.Different dose-and time-dependent effects of butyric acid on the physiological and pathological functional regulation of hUC-MSCs are different.
