摘 要:目的探寻鼠李糖乳杆菌GG(LGG)上清液在肠上皮细胞中是否通过表皮生长因子受体(EGFR)调控5-羟色胺转运体(SERT)的表达,以明确LGG治疗IBS的机制。方法分别用LGG上清超滤液、EGF、LGG上清超滤液+AG1478、EGF+AG1478等干预Caco-2细胞和HT-29细胞12 h和24 h,采用定量逆转录聚合酶链反应(RT-qPCR)和Western blotting检测EGFR及SERT表达水平。结果LGG上清超滤液和EGF干预Caco-2细胞和HT-29细胞12 h和24 h,EGFR和SERT表达水平均高于对照组,差异有统计学意义(均P<0.05)。当在Caco-2细胞和HT-29细胞中加入AG1478抑制EGFR后,LGG上清超滤液和EGF在干预24 h后SERT的上调表达均受到抑制,且差异有统计学意义(均P<0.05)。结论LGG上清液可通过激活肠上皮细胞中的EGFR上调SERT表达。Objective To determine the regulatory mechanism of Lactobacillus rhamnosus GG(LGG)supernatants on SERT expression via epidermal growth factor receptor(EGFR)in intestinal epithelial cells.Methods Caco-2 cells and HT-29 cells were treated with LGG supernatants and epidermal growth factor(EGF)in the presence or absence of AG1478(EGFR kinase inhibitor)for 12 h and 24 h,respectively.In addition,the levels of SERT mRNA and EGFR mRNA were detected by quantitative reverse transcription polymerase chain reaction(RT-qPCR).The expressions of SERT and EGFR proteins were detected by Western blotting.Results The expressions of EGFR and SERT were also higher in LGG supernatant ultrafiltrate group and EGF group compared with the control group in Caco-2 cells after 12 h and 24 h(all P<0.05).However,blocking EGFR by AG1478 could abolish the effects of LGG supernatant ultrafiltrate and EGF on up-regulation of SERT mRNA and SERT protein in Caco-2 after 24 h(all P<0.05).The similar results was found in the HT-29 cells.Conclusion LGG supernatants can up-regulate mRNA and protein expression levels of SERT via activation of EGFR in intestinal epithelial cells.
