摘  要:目的利用网络药理学的方法,建立沙参麦冬汤的活性成分作用靶点通路的网络图,对作用靶点的基因功能和通路进行富集分析,从而探讨沙参麦冬汤治疗干眼的可能作用机制。方法通过中药系统药理学数据库与分析平台(TCMSP)及中医药综合数据库(TCMID)收集沙参麦冬汤药物有效成分,通过GeneCards数据库收集干眼靶点,将有效成分靶点与干眼靶点取交集,利用GeneCards数据库和OMIM数据库去重、合并,预测沙参麦冬汤的相关靶标,筛选沙参麦冬汤的活性成分治疗疾病的潜在作用靶标。利用String平台进行蛋白质的相互作用分析,构建并挖掘PPI网络中潜在的蛋白质功能模块。采用Metascape平台分析“药物-成分-疾病-靶点”及其参与的生物过程及通路,而后采用Cytoscape 3.8.2软件构建“沙参麦冬汤成分-干眼靶点-通路”网络。凭借蛋白质相互作用数据库(STRING)创建潜在靶标蛋白的相互作用网络,并找到沙参麦冬汤治疗干眼的关键基因蛋白。使用Metascape对沙参麦冬汤治疗干眼的潜在作用靶标进行生物学功能和代谢通路分析。结果沙参麦冬汤中共筛选出163种有效活性成分,核心靶点有白蛋白(ALB)、AKT1、白介素(IL-6)、VEGFA、MAPK8、CASP3、EGFR、PPAGR等,沙参麦冬汤治疗干眼的生物学通路主要作用于TNF signaling pathway信号通路、cAMP signaling pathway信号通路、Retinol metabolism信号通路等,其功能主要是对炎性反应信号通路关键蛋白的调控作用,促进眼表上皮细胞和杯状细胞分泌黏蛋白。结论研究初步揭示了沙参麦冬汤治疗干眼的多成分、多靶点、多通路的作用机制,为沙参麦冬汤的临床开发利用提供基础。Objective In order to explore the possible mechanism of action of Shashen Maidong Decoction(沙参麦冬汤)in the treatment of dry eye,the network diagram of target pathway of the active components of Shashen Maidong Decoction was es-tablished by means of network pharmacology,and the gene function and pathway of target pathway were enriched and analyzed.Methods The effective components of Shashen Maidong Decoction were collected through TCMSP and TCMID,and the dry eye targets were collected through GeneGards database.The intersection of the effective component targets and dry eye targets was ob-tained.GeneCards database and OMIM database were used to deweighting and merge,to predict the related targets of Shashen Maidong Decoction,and to screen the potential targets of the active components of Shashen Maidong Decoction in the treatment of diseases.The String platform was used for protein interaction analysis,and the potential protein functional modules in PPI net-work were constructed and explored.Metascape platform was used to analyze"medicine-components-disease-target"and the biological processes and pathways involved,and then Cytoscape 3.8.2 software was used to build the network of"ingredients-dry eye target-pathway"of Shashen-Ophiopogon soup.The interaction network of potential target proteins was created with the protein Interaction Database(STRING),and the key gene proteins of shashenmaidong decoction in treating dry eye were found.Metascape was used to analyze the biological functions and metabolic pathways of potential targets of Shashen Maidong Decoction in the treatment of dry eyes.Results A total of 163 active ingredients were screened out in Shashen Meidong Decoction,and the core targets were ALB、AKT1、IL-6、VEGFA、MAPK8、CASP3、EGFR、PPAGR,etc.The biological pathways of Shashen Maid-ong Decoction in the treatment of dry eye mainly act on TNF signaling pathway,cAMP signaling pathway,retinol metabolism pathway,etc.Its function mainly regulates the key proteins in the inflammatory signaling pa
