益气通络排浊颗粒对慢性非细菌性前列腺炎大鼠锌离子、超氧化物歧化酶水平的影响

摘　　要：目的探究益气通络排浊颗粒治疗慢性非细菌性前列腺炎(CNP)的作用机制。方法选用56只健康的SPF级Wistar雄性大鼠,2月龄,随机分为空白组、模型组、中药对照组(前列解毒胶囊)、西药对照组(西乐葆)、益气通络排浊颗粒剂量组(高、中、低剂量),共7组,每组8只。选用角叉菜胶前列腺注射制备大鼠慢性非细菌性前列腺炎模型,造模成功后21 d开始给予各组大鼠相应的干预药物灌胃,连续给药3周后处死大鼠,摘取前列腺组织,采用锌离子检测试剂盒测定大鼠前列腺组织匀浆锌离子(Zn^(2+))浓度,ELISA法测定大鼠血浆超氧化物歧化酶(SOD)浓度。结果与空白组比较,模型组SOD、Zn^(2+)含量显著降低(P<0.05);与模型组相比,益气通络排浊颗粒剂量组(中)、西药对照组(西乐葆)中Zn^(2+)含量显著升高(P<0.05),益气通络排浊颗粒剂量组(高、中、低)、中药对照组(前列解毒胶囊)、西药对照组(西乐葆)中SOD含量显著升高(P<0.05)。结论益气通络排浊颗粒能够通过提升角叉菜胶CNP大鼠模型血浆SOD及前列腺组织中Zn^(2+)浓度恢复前列腺功能。Objective To explore the mechanism of Yiqi Tongluo Paizhuo Granule(益气通络排浊颗粒)in the treatment of chronic nonbacterial prostatitis(CNP).Methods Fifty-six healthy male SPF Wistar rats,2 months old,were randomly divided into 7 groups:blank group,model group,traditional Chinese medicine control group[Qianlie Jiedu Capsule(前列解毒胶囊)],western medicine control group(Celebrex)and Yiqi Tongluo Paizhuo Granule groups(high,medium and low doses),8 rats in each group.The chronic nonbacterial prostatitis model was established by injecting carrageenan into the prostate.The rats in each group were given the corresponding intervention drugs by gavage on the 21st day after successful modeling.After 3 weeks of continuous administration,the rats were killed and the prostate tissue was extracted.The zinc ion(Zn^(2+))concentration in the prostate tissue homogenate was determined by zinc ion detection kit and the superoxide dismutase(SOD)in plasma was deter-mined by ELISA SOD concentration.Results Compared with those of the blank group,the contents of SOD and Zn^(2+)in the model group were decreased significantly(P<0.05).Compared with those of the model groups,the contents of Zn^(2+)in the Yiqi Tongluo Paizhuo Granule medium dose group and Western medicine control group(Celebrex)were increased significantly(P<0.05).The contents of SOD in Yiqi Tongluo Paizhuo Granule groups(high,medium and low doses),traditional Chinese medicine control group(Qianlie Jiedu Capsule)and Western medicine control group(Celebrex)were increased significantly(P<0.05).Conclu-sion Yiqi Tongluo Paizhuo Granule can improve the concentration of SOD in plasma and Zn^(2+)in the prostate tissue of carrageenan CNP rats.
