摘  要:目的利用乙醇造成胃黏膜上皮细胞(GES-1细胞)损伤模型,探讨高良姜、红豆蔻、大高良姜3味山姜属中药的挥发油对GES-1细胞损伤的保护作用及作用机制。方法分别以含0~9%乙醇浓度的培养液作用于GES-1细胞,作用时间分别为2、3、4、5、6 h,筛选最佳造模条件;利用乙醇造成GES-1细胞损伤模型,测定经3味山姜属中药挥发油含药培养基处理后的细胞存活率;ELISA法测定细胞上清液中的白细胞介素8(IL-8)、肿瘤坏死因子α(TNF-α)和血管内皮生长因子(VEGF)水平;Western-blot法检测各实验组中碱性成纤维细胞生长因子(bFGF)蛋白的表达量。结果6%乙醇溶液处理3 h为造成GES-1细胞损伤的最佳造模条件;与模型组比较,3味山姜属中药的挥发油低、中、高剂量组均能显著升高GES-1细胞存活率(P<0.01);经各药挥发油处理后,细胞上清液的IL-8和TNF-α水平明显降低,VEGF水平显著升高,bFGF蛋白表达量升高(P<0.01或P<0.05)。结论高良姜、红豆蔻、大高良姜3味山姜属中药挥发油均能保护乙醇损伤的GES-1细胞,减轻细胞的受损程度,其作用机制可能与其降低IL-8和TNF-α水平,升高VEGF水平,上调bFGF蛋白表达水平有关。Objective The protective effect and mechanism of volatile oil from Gaoliangjiang(Rhizoma Alpiniae Officinarum),Hongdoukou(Fructus Galangae)and Dagaoliangjiang(Rhizoma Alpiniae Galangae)on the damage of GES-1 cells were studied by using the model of gastric mucosa epithelial cells(GES-1)injury induced by ethanol.Method The culture medium contai-ning 0-9%ethanol was used to act on GES-1 cells for 2,3,4,5 and 6 hours respectively,and the best modeling conditions were screened.The cell survival rate of GES-1 cells treated with volatile oil of Gaoliangjiang(Rhizoma Alpiniae Officinarum),Hongdoukou(Fructus Galangae)and Dagaoliangjiang(Rhizoma Alpiniae Galangae)was determined by ethanol-induced cell in-jury model.The levels of interleukin-8(IL-8),tumor necrosis factor-α(TNF-α)and vascular endothelial growth factor(VEGF)secreted in cell supernatant were measured by ELISA.Western-blot method was used to detect the expression of basic fibroblast growth factor(bFGF)protein in each test group.Result The treatment with 6%ethanol solution for 3 h was the best modeling condition for GES-1 cell injury.Compared with the model group,the low,medium and high dose groups of the volatile oil of Gaoliangjiang(Rhizoma Alpiniae Officinarum),Hongdoukou(Fructus Galangae)and Dagaoliangjiang(Rhizoma Alpiniae Galangae)can significantly increase the survival rate of GES-1 cells(P<0.01).After treatment with the volatile oil of each drug,the levels of IL-8 and TNF-αin the cell supernatant were significantly reduced,the level of VEGF was significantly in-creased and the expression of bFGF protein increased(P<0.01 or P<0.05).Conclusion The volatile oil of Gaoliangjiang(Rhi-zoma Alpiniae Officinarum),Hongdoukou(Fructus Galangae)and Dagaoliangjiang(Rhizoma Alpiniae Galangae)can protect GES-1 cells injured by ethanol and reduce the degree of cell damage.The mechanism may be related to the decrease of levels of IL-8 and TNF-α,the increase of VEGF levels and the up regulation of bFGF protein expression.
