摘　　要：目的建立气道炎性反应模型豚鼠血浆中羟基廿碳四烯酸(HETEs)的测定方法,以此研究野鸢尾黄素降低气道炎性反应的作用机制。方法以格列苯脲为内标,采用Triton X-114对血浆样品进行浊点萃取,结合LC-MS/MS法MRM监测模式测定气道炎性反应模型豚鼠血浆中HETEs的浓度。结果血浆中(±)5-HETE、(±)15-HETE在1~100 ng·mL^(-1)内,(±)12-HETE在25~2.5×10^(3)ng·mL^(-1)内线性关系良好,日内精密度为5.01%~12.24%,日间精密度为5.19%~12.59%,批内准确度89.39%~107.38%,批间准确度88.09%~104.84%。结论该方法环保、准确、稳定,可用于豚鼠血浆中HETEs的测定。Objective To establish a method for the determination of hydroxy-eicosatetraenoic acid(HETEs)in plasma of guinea pigs with airway inflammation model,and to investigate the effect of Irigenin on reducing airway inflammatory response.Method The concentration of HETEs in guinea pig plasma with airway inflammation was determined by LC-MS/MS with MRM monitoring mode,using Triton X-114 as the cloud point extraction and Glibenclamide as internal standard.Results The linearity of(±)5-HETE、(±)15-HETE in the range of 1-100 ng·mL^(-1) was good,the linearity of(±)12-HETE in the range of 25-2.5×10^(3)ng·mL^(-1)was also good,the inter-day precision was 5.01~12.24%,the intra-day precision was 5.19~12.59%,the intra-assay accuracy was 89.39~107.38%,the inter-assay precision was 88.09~104.84%.Conclusion The method is environmentally friendly,accurate and stable,and can be used for the determination of HETEs in guinea pig plasma.
