摘 要:目的:研究Notch信号通路相关受体Notch1、Notch2、Notch3、Notch4及配体Delta样典型Notch配体1、3、4(DLL1、DLL3、DLL4)、Jagged1、Jagged2在复发性流产(RSA)患者绒毛及蜕膜组织中的表达情况。方法:收集行人工流产术的RSA患者(RSA组)及正常早孕终止妊娠患者(正常组)绒毛及蜕膜组织标本各15例,并提取组织中总RNA及蛋白,实时定量PCR(qRT-PCR)及蛋白免疫印迹(Western blot)法检测两组绒毛及蜕膜组织中Notch1、Notch2、Notch3、Notch4及DLL1、DLL3、DLL4、Jagged1、Jagged2在mRNA及蛋白水平的表达情况。结果:与正常组相比,RSA组绒毛中Notch1、Notch2、Notch4、DLL4、Jagged1、Jagged2在mRNA及蛋白水平表达均降低(均P<0.01),Notch3、DLL1、DLL3在两组间表达水平比较无统计学差异(均P>0.05);与正常组相比,RSA组蜕膜中Notch1、Notch2、Notch4、DLL1、DLL4、Jagged1、Jagged2在mRNA及蛋白水平表达均降低(均P<0.01),Notch3、DLL3在两组间表达水平表达比较无统计学差异(均P>0.05)。结论:Notch信号通路部分相关受体及配体在RSA患者绒毛及蜕膜组织中呈现病理性差异表达,提示Notch信号通路与RSA发病相关,从妊娠建立伊始即参与了RSA发生。Objective:To study the expression of Notch signaling pathway-related receptors Notch1,Notch2,Notch3,Notch4 and ligand Delta like typical notch ligands 1,3,4(DLL1,DLL3,DLL4),Jagged1 and jagged2 in the villi and decidua of patients with recurrent spontaneous abortion(RSA).Methods:Fifteen samples of villi and decidua from patients with RSA(RSA group)and patients with normal early pregnancy termination(normal group)who underwent induced abortion were collected.The total RNA and protein in the tissues were extracted.The mRNA and protein levels of Notch1,Notch2,Notch3,Notch4,DLL1,DLL3,DLL4,Jagged1 and Jagged2 in the villi and decidua of the two groups were detected by qRT-PCR and Western blot.Results:Compared with the normal group,the mRNA and protein levels of Notch1,Notch2,Notch4,DLL4,Jagged1 and Jagged2 the in the villi of the RSA group were decreased(all P<0.01),while the mRNA and protein levels of Notch3,DLL1 and DLL3 had no difference between the two groups(all P>0.05).Compared with the normal group,the mRNA and protein levels of Notch1,Notch2,Notch4,DLL1,DLL4,Jagged1 and Jagged2 in the decidua of the RSA group were decreased(all P<0.01),while the mRNA and protein levels of Notch3 and DLL3 had no difference between the two groups(all P>0.05).Conclusion:Some receptors and ligands related to the Notch signaling pathway are pathologically differentially expressed in the villi and decidua of RSA patients,suggesting that the Notch signaling pathway is associated with the pathogenesis of RSA and involved in the occurrence of RSA from the beginning of pregnancy.
