摘 要:miRNA在原发性高血压(essential hypertension,EH)的发生发展中具有重要的调控功能,已成为EH新的生物标记物及治疗靶点。EH中医证候的形成、发展及转归与血清miRNA表达的特异性和时序性特征相似,以miRNA为EH中医证候研究的切入点具有重要意义,特别是为其中医证候要素的生物标志物研究及探索二者的相关性提供了理论支撑。但现有研究仅停留在通过序列分析预测与EH病理生理机制相关的miRNAs,并通过实验进行相关性验证,而miRNAs具体通过何种信号通路作用于神经机制及肾素-血管紧张素-醛固酮系统(renin-angiotensin-aldosterone system,RAAS)从而引发EH仍需借助生物信息学技术进一步挖掘;第二,部分miRNAs已经被证实为EH的新型治疗靶点,但研究仅停留在理论层面,尚未有学者提出基于miRNAs治疗EH的相关治疗方案,今后应重视以miRNAs为靶点结合现代医学及分子生物学技术服务于临床。此外,miRNA与EH中医证候相关性的研究较少,多局限于筛选特有miRNAs,EH中医证候要素形成的具体生物学标志物尚不明确,今后应积极寻找能够指导EH中医辨证分型的有效生物标志物,并将其定量化和标准化。miRNAs have important regulatory functions in the occurrence and development of essential hypertension(EH),and it has become new biomarkers and therapeutic targets for EH.The formation,development,and outcome of TCM syndromes in EH are similar to the specificity and temporal characteristics of serum miRNA expression.It is of great significance to use miRNA as an entry point for the study of TCM syndromes in EH,especially as a biomarker for TCM syndrome elements.It provides theoretical support for the correlation between physical research and exploration.However,the existing research only stops at predicting the miRNAs related to the pathophysiological mechanism of EH by sequence analysis,and verifying the correlation through experiments,and what signaling pathway specifically affects the neural mechanism and the renin-angiotensin-aldosterone system of miRNAs(RAAS)to trigger EH still needs to be further explored with the help of bioinformatics technology;secondly,some miRNAs have been confirmed as new therapeutic targets for EH,but the research is only at the theoretical level,and there are no scholars yet.The related treatment plans based on miRNAs for the treatment of EH are proposed.In the future,attention should be paid to the use of miRNAs as targets combined with modern medical and molecular biology techniques to serve the clinic.In addition,there are few studies on the correlation between miRNAs and TCM syndromes of EH,and most of them are limited to the screening of unique miRNAs.The specific biological markers for the formation of TCM syndrome elements in EH are still unclear.In the future,we should actively search for effective biomarkers that can guide EH TCM syndrome differentiation,quantify them and standardize them.
