益气活血解毒方对胰腺癌晚期胰腺细胞迁移和侵袭的影响

摘　　要：目的:探讨分析益气活血解毒方对胰腺癌晚期胰腺癌细胞迁移和侵袭的作用及其机制。方法:选取人胰腺癌PANC-1细胞及25只4周龄左右健康清洁级雌性裸鼠,随机分为观察组、化疗组及益气活血解毒方低、中、高浓度组。细胞及裸鼠处理完成后,Transwell小室实验观察各组胰腺癌细胞迁移和侵袭情况;Western Blot检测胰腺癌细胞中丝氨酸/苏氨酸激酶30(serine/threonine kinase 30,STK30)及成纤维激活蛋白β(fibroblast activating proteinβ,FAB-β)的蛋白表达水平;流式细胞仪检测裸鼠CD 4^(+)CD_(25)^(+)T细胞的比例。结果:细胞处理4 h时,与观察组比较,益气活血解毒方中、高浓度组迁移和侵袭细胞数量显著降低(P<0.05)。细胞处理6 h时,与观察组比较,各治疗组迁移和侵袭细胞数量显著降低(P<0.05);与化疗组比较,益气活血解毒方高浓度组迁移和侵袭细胞数量明显降低(P<0.05)。与观察组比较,益气活血解毒方中、高浓度组STK30、FAB-β蛋白表达水平显著降低(P<0.05);且益气活血解毒方中、高浓度组大鼠血清中CD 4^(+)CD_(25)^(+)T细胞比例明显升高(P<0.05)。结论:益气活血解毒方可提高化疗后胰腺癌大鼠免疫功能,降低胰腺癌晚期胰腺细胞的迁移和侵袭,其作用与降低胰腺癌细胞中SKT30、FAB-β蛋白表达水平有关。Objective:To investigate and analyze the effect of Yiqi Huoxue Jiedu Formula on immune function of pancreatic cancer cells after advanced chemotherapy and its effect on migration and invasion of pancreatic cancer cells and its mechanism.Methods:Human pancreatic cancer PANC-1 cells and 25 healthy and clean female nude mice about 4 weeks old were selected and randomly divided into observation group,chemotherapy group and low,medium and high concentration groups of Yiqi Huoxue Jiedu Formula.After the cells and nude mice were treated,the migration and invasion of pancreatic cancer cells in each group were observed by Transwell assay;Western Blot was used to detect Serine/Threonine Kinase 30(STK30)and fibroblast activation protein in pancreatic cancer cellsβ(fibroblast activating proteinβ,FAB-β)protein expression level;flow cytometry detected the proportion of CD_(4)^(+)CD_(25)^(+)T cells in nude mice.Results:When the cells were treated for 4 hours,compared with the observation group,the numbers of migrating and invasive cells in the middle and high concentration groups of Yiqi Huoxue Jiedu Fang were significantly decreased(P<0.05).When the cells were treated for 6 h,compared with the observation group,the number of migrating and invasive cells in each treatment group was significantly decreased(P<0.05);compared with the chemotherapy group,the number of migrating and invasive cells in the high-concentration Yiqi Huoxue Jiedu Formula group was significantly decreased(P<0.05).Compared with the observation group,the protein expression levels of STK30 and FAB-βin the middle and high concentration groups of Yiqi Huoxue Jiedu Formula were significantly decreased(P<0.05);The proportion of cells was significantly increased(P<0.05).Conclusion:Yiqi Huoxue Jiedu Formula can improve the immune function of pancreatic cancer rats after chemotherapy,reduce the migration and invasion of pancreatic cells in advanced pancreatic cancer,and its effect is related to reducing the expression of SKT30 and FAB-βin pancreatic cancer cells
