摘  要:目的:观察巴戟天寡糖(MOO)对去卵巢大鼠心肌缺血再灌注损伤(IRI)后心肌能量代谢障碍和细胞结构损伤的预防作用。方法:75只健康雌性SD大鼠按随机数字表法分为5组,对照组、假IRI组、IRI组、高剂量MOO组[2.1 g/(kg·d)]和低剂量MOO组[0.7 g/(kg·d)],每组15只。对照组仅开腹并游离卵巢,假IRI组开腹切除双侧卵巢并于7 d后开胸暴露心脏,其余3组均按去卵巢+心肌IRI模型处理,其中低、高剂量MOO组均在心肌IRI前给予相应剂量MOO药液灌胃,1次/d,连用7 d。实验结束后取出心脏,采用高效液相色谱法检测心肌组织中三磷酸腺苷(ATP)、二磷酸腺苷(ADP)和一磷酸腺苷(AMP)等高能磷酸化合物水平,计算心肌总腺嘌呤核苷酸(TAN)和心肌细胞能荷(EC);电镜下观察心肌组织超微结构变化。结果:与对照组和假IRI组比较,IRI组心脏组织中ATP、ADP、AMP、TAN和EC水平降低(P<0.05)。与IRI组比较,低、高剂量MOO组ATP、ADP、AMP、TAN和EC水平升高,高剂量MOO组以上指标高于低剂量MOO组(P<0.05)。电镜结果显示,高剂量MOO组心肌超微结构仅可见轻微损伤,损伤程度小于IRI组;低剂量MOO组心肌超微结构损伤轻于IRI组,但较高剂量MOO组严重。结论:MOO能够通过改善心肌细胞能量代谢,减轻去卵巢大鼠心肌IRI的程度。Aim:To investigate the preventive effects of morinda officinalis oligose(MOO)on energy metabolism disorder and cell structure injury of myocardium in ovariectomized rats after myocardial ischemia-reperfusion injury(IRI).Methods:Seventy-five female SD rats were randomly allocated into control group(n=15),sham IRI group(n=15),IRI group(n=15),high-dose MOO group[2.1 g/(kg·d)](n=15)and low-dose MOO group[0.7 g/(kg·d)](n=15).The rats in control group received the open abdomen operation and ovarian separation;the rats in the sham IRI group received bilateral ovariectomy first and thoracotomy after 7 days;the rats in the other 3 groups were made myocardial IRI models after ovariectomy.Before IRI,the rats of the two MOO groups were given MOO by gavage,once per day,for 7 days.After the treatment,the cardiac tissue was prepared.High energy phosphate compounds such as adenosine triphosphate(ATP),adenosine diphosphate(ADP)and adenosine monophosphate(AMP)were detected by high performance liquid chromatography,and total adenine nucleotide(TAN)and energy charge(EC)were calculated.The ultrastructural changes of myocardium were observed under electron microscope.Results:Compared with control group and the sham IRI group,ATP,ADP,AMP,TAN and EC levels in myocardial tissue of the IRI group were significantly decreased(P<0.05).Compared with the IRI group,the above indexes in the low-,high-dose MOO groups were signigicantly increased,especially in the high-dose MOO group(P<0.05).The ultrastructure of myocardium in the high-dose MOO group was only slightly injured,which was significantly improved compared with the IRI group;the injury of myocardial cells in the low-dose MOO group was milder than that in the IRI group,but more serious than that in the high-dose MOO group.Conclusion:MOO can alleviate myocardial IRI in the ovariectomized rats by improving myocardial energy metabolism.
