摘 要:研究以低共熔溶剂结合超声辅助萃取技术提取番石榴叶总黄酮。以番石榴叶为原料,低共熔溶剂为溶媒,在单因素试验基础上,分析低共熔溶剂组成、氢键受体和供体的物质的量的比、液料比、提取时间对番石榴叶总黄酮提取率的影响,利用Box-Behnken原理设计响应面试验,优化提取工艺参数。结果显示:低共熔溶剂氯化胆碱-乙二醇效果较其他溶剂效果好,物质的量的比1∶15、提取时间45 min、液料比17 mL/g是最佳提取参数,得到总黄酮提取率为21.01%,相对标准偏差(RSD)为1.1%,与回归模型预测值相差0.98%。研究表明,以氯化胆碱-乙二醇体系的低共熔溶剂可用于番石榴叶总黄酮的提取,经响应面优化的工艺提取效果良好。In the study,a highly and efficient extraction technique for determination of flavonoids from guava leaves using a combination of ultrasound-assisted extraction and deep eutectic solvents(DES)was developed.Using guava leaves as raw materials and deep eutectic solvents as solvent,response surface experiments were designed based on Box-Behnken principle.The effect of deep eutectic solvents composition,the ratio of moles of hydrogen bond acceptor and donor,liquid-solid ratio and extraction time on extraction yield of total flavonoids from guava leaves were analyzed,and the extraction parameters were optimized.The results showed that deep eutectic solvents choline chloride/ethylene glycol was better than other solvents.The ratio of moles 1∶15,extraction time 45 min and liquid-solid ratio 17 mL/g were the best extraction parameters.The extraction yield of total flavonoids was 21.01%,RSD was 1.1%,0.98%different from the predicted value of regression model.The study indicates that the deep eutectic solvents of choline chloride/ethylene glycol system can be used for the extraction of total flavonoids from guava leaves,and the extraction effect is good with the optimization of forward response surface.
