摘 要:[目的]建立一种混合多个PCR产物的高效单色毛细管电泳技术。[方法]以杂交水稻丰两优3305和常规稻润稻118为试验材料,结合品种指纹数据和PCR产物片段大小之间的差异,筛选出适用于该品种纯度鉴定的SSR分子标记。通过单重和多重毛细管电泳结果比对,验证多重电泳技术应用于SSR分子标记检测水稻纯度的实用性。[结果]片段差异10 bp及以上不同分子标记的PCR产物可以进行多重电泳。多重毛细管电泳技术既能降低检测成本,又能提高试验效率,可进一步提升毛细管电泳仪使用效能。[结论]该研究可为高效检测水稻种子纯度提供技术参考。[Objective]To establish a high-efficiency single-color capillary electrophoresis technology for mixing multiple PCR products.[Method]The hybrid rice Fengliangyou 3305 and the conventional rice Rundao 118 were used as experimental materials,based on the difference between cultivar fingerprint data and PCR product fragment size,to screen the SSR molecular markers suitable for the purity identification of this variety.Comparison of singleplex and multiplex capillary electrophoresis results,to verify the practicability of multiplex electrophoresis technology applied to SSR molecular markers to detect the purity of rice.[Result]The results showed that PCR products with different molecular markers with a fragment difference of 10 bp or more can be multiplex electrophoresed.Multiple capillary electrophoresis technology can not only reduce the detection cost but also improve the experimental efficiency,it can further improve the performance of the capillary electrophoresis instrument.[Conclusion]The study can provide a technical reference for the efficient detection of rice seed purity.
