摘 要:目的探讨不同的热暴露方式对大鼠生理功能、海马神经元形态及凋亡相关蛋白表达的影响。方法选用雄性SD大鼠30只,随机分为对照组、热暴露组和脱水式热暴露组,每组10只。热暴露后,检测各组大鼠的食物消耗量、血压及心率;尼氏染色观察海马神经元损伤情况;免疫组织化学法检测Bax、Bcl-2阳性细胞表达数目,Western Blot检测海马组织凋亡相关蛋白Bax、Bcl-2、caspase-3的表达情况。结果与对照组相比,热暴露组和脱水式热暴露组的食物消耗量均降低(P均<0.05),脱水式热暴露组低于热暴露组(P<0.05);两个热暴露组的血压和心率高于对照组(P均<0.05),且脱水式热暴露组变化大于热暴露组(P<0.05);与对照组相比,热暴露组大鼠海马神经元形态受损,数量减少(P<0.05),凋亡相关蛋白Bax、caspase3表达上升且Bax阳性细胞率增加(P均<0.05),Bcl-2表达下降且阳性细胞率减少(P<0.05),而脱水式热暴露组的上述变化较热暴漏组更加明显(P均<0.05)。结论热暴露可引起大鼠海马神经元损伤,并具有促凋亡作用,脱水式热暴露会加重这一改变。Objective To explore the effects of different heat exposure methods on the physiological function,the morphology of hippocampal neurons and the expression of apoptosis-related proteins in rats.Methods Thirty male SD(Sprague Dawley)rats were randomly divided into control,heat exposure,and dehydration heat exposure group(n=10 in each group).After a week of heat exposure,the food consumption,blood pressure and heart rate of rats in each group were measured after 7 days.The damage to hippocampal neurons was observed by Nissl staining.The number of Bax and Bcl-2 positive cells was detected by immunohistochemistry,and the expression of apoptosis-related proteins Bax,Bcl-2,and caspase-3 was detected by Western blot.Results Compared with the control group,the food consumption of the two heat exposure groups decreased(P all<0.05),and the dehydration heat exposure group was lower than that of the heat exposure group(P<0.05).The blood pressure and heart rate of the two heat exposure groups were significantly higher than the control group(P all< 0.05),and the dehydration heat exposure group aggravated the changes of the heat exposure group(P<0.05).Compared with the control group,the morphology of hippocampal neurons in the heat exposure group was damaged,the number decreased(P<0.05),the expression of apoptosis-related proteins Bax and caspase-3 increased and the positive cell rate of Bax increased(P all<0.05),while the expression of Bcl-2 was decreased and the positive cell rate decreased(P<0.05),while the above-mentioned changes were more significant in the dehydration heat exposure group(P all<0.05).Conclusion Heat exposure can cause damage to hippocampal neurons in rats,and it can obviously promote apoptosis.Dehydrated heat exposure can aggravate this change.
