聚赖氨酸/细菌纤维素抗菌敷料对烫伤大鼠创面菌群、胶质细胞活性及TGFβ1蛋白的影响

摘　　要：目的聚赖氨酸/细菌纤维素抗菌敷料对烫伤大鼠创面菌群、胶质细胞活性及TGFβ1蛋白的影响。方法选取健康雄性SD大鼠40只,将其分为分为健康组、烫伤组、PLL/BNC组、药物对照组,10只/组,除健康组10只大鼠外,其余均建立烫伤模型,于建模后第2天烫伤组用氯化钠溶液纱布外敷包扎;药物对照组用无机诱导活性敷料软膏敷于创面并包扎;PLL/BNC组采用聚赖氨酸/细菌纤维素抗菌敷料敷于创口处,均每日更换一次,连续干预14d。检测并观察各组大鼠创面组织溶菌酶表达及愈合情况,HE染色观察病理组织表达,免疫组化法检测星形胶质细胞标志物胶质细胞源性神经营养因子(GDNF),Western-blotting检测转化生长因子β1(TGFβ1)、Ⅰ型胶原蛋白、Ⅱ型胶原蛋白表达。结果健康组皮肤组织良好;烫伤组大鼠在治疗后7d、14d溶菌环直径和溶菌酶含量均较低(P<0.05);与烫伤组大鼠比较,药物对照组大鼠在治疗后7d、14d溶菌环直径和溶菌酶含量均升高(P<0.05);与药物对照组比较,PLL/BNC组大鼠在治疗后7d、14d溶菌环直径和溶菌酶含量均升高(P<0.05)。健康组大鼠皮肤组织良好;与烫伤组比较,药物对照组治疗后7d、治疗后14d愈合增加(P<0.05);与药物对照组比较,PLL/BNC组治疗后7d、治疗后14d创面愈合增加(P<0.05)。健康组大鼠皮肤组织良好;烫伤组皮肤、毛囊坏死及炎症细胞浸润严重;药物对照组仍有坏死细胞及毛囊,有少量毛细血管生长;PLL/BNC组新生血管明显增多,细胞排列较药物对照组整齐。与健康组比较,烫伤组大鼠GDNF阳性细胞平均光密度、阳性面积均升高(P<0.05);与烫伤组比较,药物对照组大鼠以上表达均降低(P<0.05);与药物对照组比较,PLL/BNC组以上表达均降低(P<0.05)。与健康组比较,模型组大鼠皮肤组织中TGFβ1、Ⅰ型、Ⅱ型胶原蛋白降低(P<0.05);与烫伤组比较,药物对照组蛋白表达升高(P<0.05);与药物对照组比Objective To observe the effects of polylysine/bacterial cellulose antibacterial dressing on wound microflora,glial cell activity and TGFβ1 protein in scald rats.Methods Forty healthy male SD rats were selected and divided into healthy group,scald group,PLL/BNC group and drug control group(10 rats/group).Scald models were established except for the healthy group(10 rats/group),and the scald group was externally wrapped with sodium chloride solution on the 2nd day after modeling.In drug control group,inorganic induced active dressing ointment was applied to wound and bandaged.In PLL/BNC group,polylysine/bacterial cellulose antibacterial dressing was applied to the wound,which was changed once a day for 14 consecutive days.The expression of lysozyme and healing in wound tissues of rats in each group were detected and observed.HE staining was used to observe the expression of pathological tissues.Immunohistochemical method was used to detect the astrocyte marker glial cell-derived neurotrophic factor(GDNF).The expression of transforming growth factorβ1(TGFβ1),typeⅰcollagen and typeⅱcollagen was detected by Western-blotting.Results The skin tissue of healthy group was good.The diameter of lysozyme ring and lysozyme content in scald group were lower than those in scald group at 7d and 14d after treatment(P<0.05).Compared with scald group,lysozyme ring diameter and lysozyme content in drug control group were increased at 7d and 14d after treatment(P<0.05).Compared with the drug control group,the lysozyme ring diameter and lysozyme content of PLL/BNC group were increased at 7d and 14d after treatment(P<0.05).The skin tissue of healthy group was good.Compared with the scald group,the healing of the drug control group was increased at 7d and 14d after treatment(P<0.05).Compared with the drug control group,the wound healing of PLL/BNC group was increased 7d and 14d after treatment(P<0.05).The skin tissue of healthy group was good.Skin and hair follicle necrosis and inflammatory cell infiltration were serious in scal
