摘  要:目的:观察痛风颗粒对高尿酸血症模型大鼠的作用并探讨其机制。方法:通过连续给大鼠喂食腺嘌呤溶液和定制高酵母饲料10 d,同时皮下注射氧嗪酸钾溶液建立高尿酸血症大鼠模型,用全自动生化分析仪测定正常组和模型组大鼠血浆尿酸、肌酐和尿素氮水平,HE染色观察大鼠关节滑膜与肝肾组织的病理变化,ELISA检测血浆炎性细胞因子的表达水平。结果:与模型组比较,痛风颗粒能显著降低高尿酸血症模型大鼠的尿酸、肌酐和尿素氮水平(P<0.01),降低肿瘤坏死因子(tumor necrosis factor,TNF)-α、白细胞介素(Interleukin,IL)-1β、IL-4、干扰素(Interferon,IFN)-γ、IL-2、IL-17、IL-13含量(P<0.01)。结论:痛风颗粒对高尿酸血症模型大鼠有较好的治疗作用,其作用机制可能与降低炎性细胞因子相关。Objective:To observe the effect of Tongfeng granules on hyperuricemia model rats,and explore its mechanism.Methods:Rats were fed with adenine solution and customized high-yeast feed continuously for 10 days,and at the same time potassium oxazinate solution was subcutaneously injected to establish a hyperuricemia rat model.Uric acid(UA),creatinine and urea nitrogen levels of the control group and the model group were measured by automatic biochemical analyzer.The pathological changes of rat joint synovium,liver tissue and kidney tissue were observed through HE staining.Elisa was used for testing the expression levels of blood inflammatory cytokines.Results:Compare with the model group,Tongfeng granules can significantly reduce the UA,creatinine and urea nitrogen levels of the hyperuricemia rat model(P<0.01),and reduce TNF-α,IL-1β,IL-4,INF-γ,IL-2,IL-17 and IL-13 contents(P<0.01).Conclusion:Tongfeng granules have shown a good therapeutic effect on a hyperuricemia rat model.Its mechanism might be related to its ability to reduce inflammatory cytokines.
