摘 要:目的:探究风流草黄酮对胫骨骨折模型大鼠骨折愈合过程中骨质变化的影响。方法:将300只6周龄SD大鼠按照给药周期(4,7,14,21,28 d)随机分为5大组,每大组60只。再将每大组按处理方式分为对照组、模型组、风流草黄酮低剂量组(15 mg/kg)、中剂量组(30 mg/kg)以及高剂量组(60 mg/kg)各12只,分别于给药4、7、14、21、28 d后用Elisa法测定相应组别大鼠胫骨中碱性磷酸酶(alkaline phosphatase,ALP)活性、总胶原蛋白及羟脯氨酸(hydroxyproline,Hyp)含量,放射免疫法测定骨钙素(osteocalcin,OCN)含量,电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry,ICP-MS)测定骨钙、骨磷含量,并比较组间差异。结果:与对照组比较,模型组不同时间点的骨代谢相关参数均有所降低(P<0.01)。与模型组比较,风流草黄酮不同剂量组不同时间点的骨代谢相关参数均有所升高(P<0.05,P<0.01),且呈时间、剂量依赖性。结论:风流草黄酮可改善胫骨骨折大鼠的骨代谢状况,促进骨形成,有助于骨折愈合。Objective:To explore the effect of Desmodium gyrans(L.)DC.flavonoids on bone changes during fracture healing in tibial fracture model rats.Methods:According to different administration time(4,7,14,21,28 days),a total of 3006-week-old SD rats were randomly divided into five large groups,with 60 rats in each large group.Then,according to different treatment methods,each large group was divided into control group,model group,and low dose,medium dose,high dose(15,30,60mg/kg)of Desmodium gyrans(L.)DC.flavonoids group,with 12 rats in each group.On the 4^(th) day,7^(th) day,14^(th) day,21^(st) day and 28^(th) day,tibia osseous routine parameters of corresponding groups of rats were tested and compared respectively.Alkaline phosphatase(ALP)activity,total bone collagen and hydroxyproline(Hyp)contents were determined by Elisa.The content of osteocalcin(OCN)was determined by radioimmunoassay.Bone calcium and phosphorus contents were determined by inductively coupled plasma mass spectrometry(ICP-MS).Results:Compared with control group,osseous routine parameters in the model group were significantly decreased(P<0.01).However,osseous routine parameters changes in the Desmodium gyrans(L.)DC.flavonoids groups significantly increased and presented time and dose dependence,compared with the model group(P<0.05,P<0.01).Conclusion:Desmodium gyrans(L.)DC.flavonoids can play a positive role in bone metabolism and osteogenesis of tibia fracture model in SD rat,which suggest it can be benefit for fracture healing.
