副猪嗜血杆菌血清7型菌株的分离鉴定及生物学特性研究

摘　　要：为了掌握河南某规模化猪场保育猪发病猪群的感染病原,研究从濒死期病猪的肺脏、心血样品中分离获得一株细菌,通过对该株细菌分离鉴定、血清型分型、致病性试验、毒力基因检测和药敏试验等一系列研究工作。结果表明,该株细菌为副猪嗜血杆菌,血清型为7型,与副猪嗜血杆菌vHPS7株(GenBank登录号:CP049089)同源性为100%。该菌株同时携带有vta1、vta2、vta3、wza、ompP2、nanH、cdtA、cdtB、dtC、espP等毒力基因,结合豚鼠致病性试验结果和临床发病情况,该菌株具有较强的致病性;对头孢噻呋、头孢他啶、强力霉素、土霉素、四环素、阿奇霉素6种药物敏感;对青霉素G、阿莫西林、氨苄西林、复方新诺明、左氟沙星、氧氟沙星、恩诺沙星、环丙沙星、壮观霉素、卡那霉素、阿米卡星11种药物耐药。该研究为今后副猪嗜血杆菌血清7型致病机制研究奠定了基础,为副猪嗜血杆菌血清7型的临床防控提供了理论基础。In order to understand the pathogen of infection in a large-scale pig farm in Henan Province,a strain of bacteria was isolated from the lung and heart blood samples of dying pigs.Through a series of research work,such as isolation and identification,serotype identification,pathogenicity test,virulence gene detection and drug sensitivity test of the strain.The results showed that the strain was Haemophilus parasuis,serotype 7,and had 100% homology with Haemophilus parasuis vhps7 strain(GenBank No.:CP049089).It carried virulence genes such as vta1,vta2,vta3,wza,ompP2,nanH,cdtA、cdtB、cdtC、espP,showed that the strain was highly pathogenic according to the results of pathogenicity test and clinical conditions of guinea pigs;the strain was sensitive to Ceftiofurr,Ceftazidime,Doxycycline,Oxytetracycline,Tetracycline and Azithromycin;it was resistant to Penicillin G,Amoxicillin,Ampicillin,Sulfamethoxazole,Levofloxacin,Ofloxacin,Enrofloxacin,Ciprofloxacin,Spectinomycin,Kanamycin and Amikacin.This study laid a foundation for the pathogenic mechanism of Haemophilus parasuis serotype 7 in the future,and provided a theoretical basis for the clinical prevention and control of Haemophilus parasuis serotype serotype 7.
