摘　　要：目的:探讨不同煎煮方法及时间对炮附子配伍大黄治疗阳虚型便秘的效果及对心脏的影响。方法:选健康成年SD大鼠,采用随机数字表法分为单煎组、合煎组与空白对照组,并进一步按煎煮时间分为5个亚组,每亚组7只。单煎组各亚组分别给予煎煮60、90、120、150、180min的合并水煎液,合煎组中各亚组分别给予煎煮60、90、120、150、180min的合煎液,空白对照组给予等体积的0.9%氯化钠溶液。观察给药后大鼠排便情况(首粒排便时间、24h内排便颗粒数);检测给药后的血清胃泌素(GAS)及血浆胃动素(MTL)、血管活性肠肽(VIP)、内皮素(ET)水平;检测给药后的大鼠心功能[左室收缩压(LVSP)、左室舒张末期压(LVEDP)、左室内最大下降速率(-dp/dt_(max))、左室内最大上升速率(+dp/dt_(max))]变化情况;观察心律失常发生情况。结果:单煎组、合煎组各亚组的首粒排便时间均短于空白对照组(P<0.05),24h内排便颗粒数均大于空白对照组(P<0.05),GAS、MTL、VIP、ET、LVSP、+dp/dt_(max)水平均高于空白对照组(P<0.05),-dp/dt_(max)、LVEDP水平均低于空白对照组(P<0.05)。合煎组各亚组的首粒排便时间均短于单煎组(P<0.05),24h内排便颗粒数均大于单煎组(P<0.05),GAS、MTL、VIP、ET、LVSP、+dp/dt_(max)水平均高于单煎组(P<0.05),-dp/dt_(max)、LVEDP水平均低于单煎组(P<0.05)。单煎组及合煎组组内煎煮120min亚组的首粒排便时间均短于同组内煎煮60min亚组、90min亚组(P<0.05),24h内排便颗粒数均大于同组内煎煮60min亚组、90min亚组(P<0.05),GAS、MTL、VIP、ET、LVSP、+dp/dt_(max)水平均高于同组内煎煮60min亚组、90min亚组(P<0.05),-dp/dt_(max)、LVEDP水平均低于同组内煎煮60min亚组、90min亚组(P<0.05)。单煎组及合煎组组内煎煮120min亚组的首粒排便时间均稍短于同组内煎煮150min亚组、180min亚组,24h内排便颗粒数均稍大于同组内煎煮150min亚组、180min亚组,GAS、MTL、VIP、EObjective:To study the effect of different decocting methods and time on the treatment of constipation of Yang defi ciency with compatibility of Processed Radix Aconiti Lateralis Praeparata with Radix et Rhizoma Rhei and its infl uence on the heart.Methods:Healthy adult SD rats were selected,and they were divided into single decoction group,combined decoction group and control group by random number table method,and further divided into 5 subgroups according to the decoction time,with 7 rats in each subgroup.Each subgroup of the single decoction group was given the combined decoction of 60,90,120,150 and 180 min,and each subgroup of the combined decoction group was given the merged decoction of 60,90,120,150 and 180 min,while the control group was given equal volume 0.9%sodium chloride solution.The defecation condition of rats after administration(fi rst defecation time,number of defecation particles within 24 h)were observed,and the levels of serum gastrin(GAS),plasma motilin(MTL),vasoactive intestinal peptide(VIP)and endothelin(ET)were measured after administration,and the changes of cardiac function of rats[left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP),rate of pressure decline during myocardial relaxation(–dp/dt_(max))and rate of pressure development during myocardial contraction(+dp/dt_(max))]were measured after administration,and the occurrence of arrhythmia were observed.Results:The fi rst defecation time of each subgroup of the single decoction group and combined decoction group were shorter than those of blank control group(P<0.05),and the number of defecation particles within 24 h were greater than those in the blank control group(P<0.05),and the levels of GAS,MTL,VIP,ET,LVSP and+dp/dt_(max) were higher than those in blank control group(P<0.05),the–dp/dt_(max) and LVEDP were lower than those of the blank control group(P<0.05).The fi rst defecation time of each subgroup in the combined decoction group were shorter than those of single decoction group(P<0.05),a
