摘 要:建立超高压液相色谱-电喷雾四极杆串联静电场轨道阱高分辨质谱联用仪对北刘寄奴药材化学成分的快速定性分析研究,以ACE C_(18)色谱柱,0.1%甲酸水-乙腈为梯度洗脱相,正负离子同时采集、HCD碰撞解离获取碎片离子数据,轨道阱回旋振荡检测成分高分辨质谱离子,得到成分碎片裂解质谱离子,综合已有数据库成分数据与碎片离子裂解规律进行化学成分结构解析。鉴定出北刘寄奴64种化学成分,其中苦参碱、桃叶珊瑚苷、地黄苷A、密力特苷,京尼平苷酸、栀子苷、车叶草苷酸、夏佛塔苷、异嗪皮啶、杠柳毒苷、金合欢素11种化合物为首次从北刘寄奴药材中鉴定出,并根据其碎片离子推测其质谱裂解途径。本方法具有快速、高效检测优势,能较全面反映出北刘寄奴中的化学成分物质,为深入分离研究北刘寄奴药效物质基础及其质量标准提供准确可靠的检测方法。Ultra high pressure liquid chromatography electrospray quadrupole tandem electrostatic field high resolution mass spectrometer was developed for rapid qualitative analysis of chemical constituents from Siphonostegia chinensis Benth,ACE C_(18) column was used in this research,0.1%formic acid water acetonitrile was used as gradient elution phase.Simultaneous acquisition of positive ion mode and negative ion mode and and HCD collision dissociation was used to obtain fragment ion data,the chemical composition and structure were analyzed by integrating the existing database composition data and the fragmentation law of fragmentions,Sixty four chemical constituents were identified,Matrine;Aucubin;Rehmannioside A,Melittoside,Geniposidic acid,Geniposide,Asperulosidic acid,Schaftoside,Isofraxidin,Periplocin,Acacetin were identified from the medicinal material Siphonostegia chinensis Benth.for the first time.This method has the advantages of rapid and efficient detection,can comprehensively reflect the chemical components in Siphonostegia Chinensis Benth,and provides an accurate and reliable detection method for the in-depth separation and study of the pharmacodynamic material basis and quality standard of Siphonostegia chinensis Benth.
