摘 要:目的:通过超高效液相色谱法(UPLC)特征图谱、色度值,比较白芍与炒白芍的差异。方法:采用UPLC建立白芍药材与炒白芍的特征图谱,并采用SIMCA 14.0软件进行主成分分析(PCA)、正交偏最小二法乘-判别分析(OPLS-DA);采用色差仪获取白芍药材与炒白芍的色度值(L^(*)、a^(*)、b^(*)),获取色差范围。结果:白芍药材的UPLC特征图谱共标定15个共有峰,炒白芍的UPLC特征图谱共标定17个共有峰。PCA共提取4个主成分,累积方差贡献率为86.7%,15批白芍药材与炒白芍在OPLS-DA模型中分类聚集明显。色彩图像分析中炒白芍与白芍药材的色差值(△E^(*))为8~20,白芍药材的b^(*)_(白芍)为1~7,炒白芍的b^(*)_(炒白芍)为9~21。结论:该方法稳定、可靠、精密度高、重复性好、色彩图像分析简单直观,可量化炮制程度,并为炮制品质量控制研究提供参考。Objective:To explore the difference between Paeoniae Radix Alba(PRA)and stir-fried PRA based on ultra-performance liquid chromatograph(UPLC)fingerprints and chromatic values.Methods:UPLC fingerprints for PRA and stir-fried PRA were established,and Simca 14.0 was employed for principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA).Chromatic meter was used to measure the chromatic values(L^(*):lightness,a^(*):red-green color value,b^(*):blue-yellow color value)of the two products and further the range of color difference was calculated.Results:A total of 15 common peaks were found in UPLC fingerprints of PRA and 17 in fingerprints of stir-fried PRA.PCA yielded 4 principal components,with the cumulative variance explained of 86.7%.OPLS-DA score plot separated the PRA samples and stir-fried PRA samples.The color difference(△E^(*))between PRA and stir-fried PRA was 8-20,and b^(*)was 1-7 for PRA and 9-21 for stir-fried PRA.Conclusion:The method is stable,reproducible,and feasible,and accurate,and the color image is intuitive.Thus,it can quantify the processing degree and be used for the quality control of processed medicinal materials.
