摘 要:目的 研究单侧听觉剥夺对听觉发育关键期小鼠内耳的影响。方法 将22只出生后12天的C57BL/6小鼠随机分为正常对照组和听觉剥夺组。对听觉剥夺组进行单侧听觉剥夺,2周后解除剥夺。对正常对照组小鼠和听觉剥夺组小鼠双耳采用听性脑干反应(ABR)评估听功能,并通过免疫荧光染色对毛细胞、带状突触、螺旋神经节进行形态学观察。统计学分析听觉剥夺组小鼠的剥夺耳、未剥夺耳和正常对照小鼠在听功能和耳蜗形态学方面的差异。结果 1)听觉剥夺2周后,听觉剥夺组的剥夺耳ABR高频阈值升高,Ⅰ波潜伏期延长,与对照组小鼠相比差异有统计学意义;2)听觉剥夺组的未剥夺耳与正常对照组间ABR阈值和Ⅰ波潜伏期差异无明显差异;3)免疫荧光染色结果示听觉剥夺耳和未剥夺耳的毛细胞形态和数量未见异常;4)和对照组相比,听觉剥夺组的剥夺耳出现了带状突触数量减少,未剥夺耳与对照组之间的带状突触数量未见统计学差异,没有出现代偿性增加。结论 单侧听觉剥夺可导致听觉发育关键期小鼠剥夺耳的听功能及带状突触异常。Objective To investigate the effects of unilateral auditory deprivation on the inner ear oinf mice during critical stage of auditory development.Methods C57BL/6 mice(n=22) were randomly divided into a control and an auditory deprivation group.Auditory input was deprived in one ear in animals in the deprivation group for 2 weeks.Auditory brainstem responses(ABRs) were used to evaluate the auditory function,and morphology of hair cells,ribbon synapses and spiral ganglion cells examined by immunofluorescence staining,with differences between the control and auditory deprivation groups statistically analyzed.Results Threshold of high frequency ABRs increased with prolonged wave I latency in the auditory deprivation group as compared with the control group(P<0.05).ABR thresholds and wave I latencies in non-deprived ears were not significantly different between the auditory deprivation and control groups.Immunofluorescence staining showed no abnormality in the morphology and number of hair cells in either deprived ears or non-deprived ears.Compared with the control group,the number of ribbon synapses decreased in deprived ears,but showed no significant difference between non-delivered ears in the deprivation group and ears in the control group.Conclusion Unilateral auditory deprivation during critical stage of auditory development may lead to abnormal auditory function related to ribbon synapses disorders.
