摘 要:目的探讨北五味子多糖(Schisandra chinensis polysaccharide,SCP)对体外培养的人膀胱癌细胞T24的作用.方法采用SCP 50、100、200μg/mL 3个剂量处理体外培养的人膀胱癌细胞T24,四甲基偶氮唑蓝(MTT)检测SCP对T24细胞增殖的抑制效应,显微镜下观察细胞的形态学改变;流式细胞仪(FCM)检测SCP对细胞周期百分率的影响;Hoechst33258荧光染色法检测SCP对T24细胞凋亡的影响;Western blot(WB)法检测SCP对T24细胞中cleaved Caspase-3、cleaved Caspase-9、Bax和Bcl-2蛋白表达的影响.结果与对照组比较,SCP可显著抑制T24细胞增殖(P<0.05),促进T24细胞凋亡;FCM结果显示:与对照组比较,SCP能明显促进凋亡,增加凋亡率(P<0.05);SCP能明显降低T24细胞中Bcl-2蛋白表达,提高Bax蛋白表达,增加cleaved Caspase-3和cleaved Caspase-9的蛋白表达,与对照组比较差异显著(P<0.05).结论SCP能抑制人膀胱癌T24细胞增殖,并促进其凋亡,其机制可能与诱导细胞凋亡和调控相关基因表达有关.Objective To investigate the molecular mechanism of Schisandra chinensis polysaccharide(SCP) on human bladder cancer cell line T24 in vitro.Method Three doses of SCP with 50,100,200 μg/mL were used to treat human bladder cancer T24 cells in vitro.MTT assay was used to detect the inhibitory effect of SCP on the proliferation of T24 cells.Cell morphology was observed under microscope;Flow cytometry was used to detect the effect of SCP on cell cycle and apoptosis rate;Hoechst 33258 fluorescence staining method was used to detect the effect of SCP on apoptosis of T24 cells;The effects of SCP on protein expression of cleaved Caspase-3,cleaved Caspase-9,Bax and Bcl-2 of T24 cells were detected by Western blot(Wb).Results SCP could significantly inhibit the proliferation of T24 cells and promote the apoptosis of T24 cells compared with the control group;Flow cytometry showed that the apoptosis rate increased in the SCP treated group(P<0.05);SCP could significantly reduce Bcl-2 of T24 cells increase Bax and increase cleaved Caspase-3 and cleaved Capase-9 protein expression compared with that of control group in a dose-dependent manner, all indicators have obvious differences compared with those of control group(P<0.05).Conclusion SCP can inhibit the proliferation and promote the apoptosis of human bladder cancer T24 cells, and the mechanism may be related to inducing cell apoptosis and regulating related gene expression.
