摘  要:目的:研究参芪薏苡仁粥通过Toll样受体4(Toll Like Receptor 4,TLR4)/核因子(Nuclear Factor,NF)-κB路径改善大鼠化疗后胃肠黏膜损伤的机制。方法:选择雄性SPF级的Wistar大鼠36只,随机分为三组:模型组、药膳组、空白组。采用5-氟尿嘧啶(5-Fluorouracil,5-FU)构建大鼠化疗后胃肠黏膜损伤模型,采用逆转录聚合酶链反应(Reverse TranscriptionPolymerase Chain Reaction,RT-PCR)法检测回肠组织TLR4/NF-κB通路蛋白和肠道紧密连接蛋白信使核糖核酸(messenger Ribonucleic Acid,m RNA)含量,免疫印迹(Western blot)法检测回肠组织通路蛋白的表达,酶联免疫吸附试验(Enzyme-Linked Immunosorbent Method,ELISA)法检测血清肿瘤坏死因子(Tumor Necrosis Factor,TNF)-α、白细胞介素(Interleukin,IL)-6水平。结果:模型组和药膳组通路蛋白及肠道紧密连接蛋白mRNA均呈上调趋势,其中模型组蛋白mRNA表达水平更显著。三组间TLR4、NF-κB p65蛋白表达水平的差异无统计学意义(P>0.05);模型组与空白组NF-κB p50、靠停蛋白1(Claudin-1,CLDN1)、遮光蛋白(Occludin,OCLN)、连接粘附分子-1(Junctional Adhesion Molecule 1,JAM-1)、紧密连接蛋白-1(Zona Occludens 1,ZO-1)蛋白表达水平的差异有统计学意义(P<0.05);药膳组与空白组B细胞κ轻肽基因增强子核因子抑制因子α(NF-Kappa-B Inhibitor Alpha,IκBα)、CLDN1、JAM-1、ZO-1蛋白表达水平的差异有统计学意义(P<0.05)。各组大鼠血清TNF-α和IL-6的浓度的差异无统计学意义(P>0.05)。结论:参芪薏苡仁粥可以促进肠道紧密连接蛋白的表达,从而减轻大鼠化疗后胃肠黏膜损伤,但对通路的炎症因子TLR4、NF-κB、TNF-α和IL-6炎症因子的抑制作用尚不明确。Objective:To explore the mechanism of the Shenqi Yiyiren(参芪薏苡仁)porridge through TLR4/NF-κB pathway in improving gastrointestinal mucosal injury in rats after chemotherapy.Methods:A total of 36 Wistar rats of SPF grade were selected and randomly divided into three groups:the model group,the medicated diet group,and the blank group.The rat model of gastrointestinal mucosal injury after chemotherapy was constructed using 5-FU,the TLR4/NF-κB pathway protein and intestinal tight junction protein mRNA were detected by RT-PCR method,and the Western blot method was used to detect the pathway protein expression.ELISA method was used to detect TNF-α and IL-6 levels.Results:The pathway protein and intestinal tight junction protein mRNA in the model group and the medicated diet group showed an up-regulation trend,and the expression level of protein mRNA in the model group was more significant.There was no statistically significant difference in TLR4 and NF-κB p65 protein expression levels among the three groups(P>0.05);there were statistical differences in NF-κB p50,CLDN1,OCLN,JAM-1,and ZO-1 protein expression levels between the model group and the blank group(P<0.05);the protein expression levels of IκBα,CLDN1,JAM-1 and ZO-1 in the medicated diet group and the blank group were significantly different(P<0.05).There was no significant difference in TNF-α and IL-6 concentrations of rats in each group(P>0.05).Conclusion:The Shenqi Yiyiren porridge can promote the expression of intestinal tight junction protein,thereby reducing the gastrointestinal mucosal damage after chemotherapy in rats,but the inhibitory effect on the inflammatory factors TLR4,NF-κB,TNF-α and IL-6 are unclear.
