摘  要:目的通过TOPSIS法(technique for order preference by similarity to an ideal solution)综合评估黑种乌头(铁棒锤,Aconitum pendulum Busch.,APB)和白种乌头[甘青乌头,Aconitum tanguticum(Maxim.)Stapf.,ATS]的体外抗神经病理性疼痛作用及其作用机制。方法体外培养小鼠小胶质细胞BV2,用三磷酸腺苷(ATP)诱导体外神经疼痛模型。设空白对照组(NC)、模型对照组(MC)及APB和ATS醇提物给药组,以细胞活力、NO释放量、炎症因子IL-6、iNOS mRNA表达和P2X4、P2X7 mRNA表达等作为评价指标,探讨APB和ATS对各指标的影响。并通过TOPSIS法将各指标数据归一化,计算两者相对接近度,综合评价优势方案。结果ATP可导致BV2细胞活力下降并促进细胞上清液中NO的释放,使iNOS和IL-6 mRNA水平显著升高,而APB和ATS提取物可显著抑制上述指标变化。而且,APB和ATS对ATP诱导的P2X7 mRNA水平上调具有更明显抑制作用。TOPSIS法结果提示,ATS的相对接近度比APB更靠近理想方案NC组。结论APB和ATS均可显著抑制ATP诱导的神经病理性疼痛,且通过TOPSIS法综合分析可得,ATS在抗神经病理性疼痛方面可能更具优势。Objective To comprehensively evaluate the anti-neuropathic pain effect of the Tibetan medicine black aconite(Aconitum pendulum Busch.,APB)and white aconite[Aconitum tanguticum(Maxim.)Stapf.,ATS]and discuss their mechanism of action based on TOPSIS(technique for order preference by similarity to an ideal solution)method.Methods In vitro neuropathic pain model was established by using BV2 cells induced by 5′-adenylate triphosphate(ATP).The cells were divided into normal control group(NC),model control group(MC),APB group and ATS group.The indicators such as cell viability,nitric oxide(NO)release as well as IL-6,iNOS,P2X4,P2X7 mRNA levels were detected to evaluate the efficiencies of APB and ATS.TOPSIS method was applied to normalize the aforementioned indexes and comprehensively estimate the advantage scheme after calculating their relative proximity.Results ATP treatment could reduce the cell viability,promote NO release,and increase the levels of iNOS and IL-6 mRNA in BV2 cells,which could be attenuated by APB and ATS extracts respectively.Moreover,both of APB and ATS could reverse the elevated mRNA levels of P2X7 in BV2 cells induced by ATP.According to the results of TOPSIS analysis,the relative proximity of ATS was closer to the ideal protocol NC group than that of APB.Conclusion APB and ATS can both ameliorate ATP induced neuropathic pain significantly.ATS has more advantages in reducing neuropathic pain according to TOPSIS analysis.
