摘  要:试验旨在探究螺旋藻多糖(Spirulina platensis polysaccharide,PSP)的抗氧化作用,首先经过酶解水提法得到螺旋藻多糖,然后经过DEAE-52纤维素层析柱和Sephacryl S-200凝胶层析柱分离纯化后得到螺旋藻多糖组分1(PSP-1)。将小鼠分为3个不同剂量药物组(200、100 mg/kg·BW和50 mg/kg·BW)、猪伪狂犬病毒(PRV)组和对照组共5个组别(A、B、C、D、E组)进行试验,采用试剂盒法测定小鼠血清中一氧化氮(NO)、诱导型一氧化氮合酶(iNOS)和丙二醛(MDA)的分泌水平以及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、髓过氧化物酶(MPO)、黄嘌呤氧化酶(XOD)和谷胱甘肽过氧化物酶(GSH-Px)活性。结果表明:在使用了不同剂量的药物之后,血清中NO、MDA水平和MPO活性均极显著降低(P<0.01);iNOS、XOD活性在A组和B组均极显著降低(P<0.01);同时,血清中SOD活性均极显著升高(P<0.01);GSH-Px活性和CAT活性在A组和B组极显著升高(P<0.01),在C组显著升高(P<0.05)。由此可见,PSP-1能够缓解PRV感染小鼠引起的氧化应激,具有一定的抗氧化性,可为动物病毒性疾病的治疗提供思路。The aim of this experiment was to ex⁃plore the antioxidant effect of polysaccharides from Spirulina chinensis.In this experiment,PSP was firstly extracted by enzymatic hydrolysis of water,and then purified by DEAE 52 cellulose chromatography column and sephacryl S-200 gel chromatography column.SPF grade BALB/c mice were divided into three different dose drug groups(200,100 mg/kg·BW and 50 mg/kg·BW),PRV group and control group,five groups in total,namely A,B,C,D,E group.Levels of nitric oxide(NO),in⁃ductive nitric oxide synthase(iNOS),malondialdehyde(MDA)and activities of superoxide dismutase(SOD),catalase(CAT),myeloperoxidase(MPO),xanthine oxidase(XOD)and glutathione peroxidase(GSH Px)were determined by kit method.The results showed that the levels of NO,MDA and MPO in serum were extremely significantly decreased after different doses of drugs were used(P<0.01).iNOS,XOD in A and B groups was extremely significantly decreased(P<0.01);meanwhile,SOD levels in serum were ex⁃tremely significantly increased(P<0.01);GSH-Px and CAT were extremely significantly increased in A group and B group(P<0.01),and were significantly increased in C group(P<0.05).In conclusion,PSP-1 could alleviate the oxidative stress caused by PRV infection in mice,and has certain antioxidant activity,which provided ideas for the treatment of animal viral diseases.
