摘  要:目的探讨人参皂苷Rg3调控长链非编码RNA(long noncoding RNA,LncRNA)PXN-AS1的表达及对肺癌细胞放射敏感性的影响。方法选用肺癌细胞株A549、H466、H1299、H522,将培养液中加入人参皂苷Rg3的肺癌细胞确定为研究组,未加药物培养的肺癌细胞为对照组。进行克隆形成实验,检测人参皂苷Rg3对肺癌细胞放射敏感性的影响。流式细胞仪检测人参皂苷Rg3对肺癌细胞凋亡的作用。通过荧光定量PCR检测肺癌组织和细胞中PXN-AS1的表达。结果与对照组比较,研究组人参皂苷Rg3能够明显降低肺癌细胞放射治疗后的克隆形成率(P<0.05),显著增加细胞凋亡(P<0.05)。PXN-AS1在肺癌组织和细胞中高表达,且与肺癌患者的预后呈负相关。加入人参皂苷Rg3后,研究组PXN-AS1的表达发生明显下调(P<0.05)。结论人参皂苷Rg3可能是潜在的放射增敏剂,可能通过调控PXN-AS1促进肺癌的放射增敏。Objective To investigate the effects of ginsenoside Rg3 regulates the expression of long non-coding RNA(LncRNA)PXN-AS1 and its effect on radiosensitivity in lung cancer cells.Methods Lung cancer cell lines A549,H466,H1299 and H522 were selected,and lung cancer cells with ginsenoside Rg3 to the culture medium were identified as the study group,and lung cancer cells cultured without drugs were identified as the control group.Clone formation assay were performed to examine the effect of ginsenoside Rg3 on radiosensitivity of lung cancer cells.The effect of ginsenoside Rg3 on the apoptosis of lung cancer cells were determined by flow cytometry.PXN-AS1 expression in lung cancer tissues and cells were determined by fluorescence quantitative PCR.Results Compared with the control group,ginsenoside Rg3 in the study group could significantly reduce the clone formation rate of lung cancer cells after radiotherapy(P<0.05),and significantly increase cell apoptosis(P<0.05).PXN-AS1 was highly expressed in lung cancer tissues and cells,and was negatively correlated with the prognosis of lung cancer patients.After the addition of ginsenoside Rg3,the expression of PXN-AS1 was significantly downregulated in the study group(P<0.05).Conclusion Ginsenoside Rg3 might be a potential radiosensitizer and may promote radiosensitization in lung cancer by regulating PXN-AS1.
