摘 要:目的:建立一个高效稳定便捷的辣椒遗传转化体系。方法:以辣椒材料BY-1的子叶作为外植体,探索影响辣椒遗传转化体系的因素,包括不同激素浓度、卡那霉素选择压、侵染时间、特美汀水溶液浸泡时间等。结果:在卡那霉素50 mg/L、农杆菌侵染时间30 min以及特美汀水溶液浸泡时间3 min下抑菌效果较好。不定芽诱导:MS+4 mg/L 6-BA+50 mg/L Kan+200μmol/L TMT+3.0%蔗糖+0.8%琼脂、不定芽伸长:MS+4 mg/L 6-BA+50 mg/L Kan+200μmol/L TMT+0.5 mg/L GA_(3)+3.0%蔗糖+0.8%琼脂,不定芽生根:1/2MS+50 mg/L Kan+200μmol/L TMT+3.0%蔗糖+0.8%琼脂。结论:得到了BY-1的遗传转化植株,并建立了该材料的遗传转化体系。Objective:To establish an efficient and convenient capsium genetic transformation system.Methods:Using Capsicum BY-1 as material and cotyledon as explants,the influencing factors of Capsicum genetic transformation system were studied,including different hormone concentration,kanamycin selection pressure,infection time,soaking time of termetine aqueous solution,etc.Results:The antibacterial effect of kanamycin 50 mg/L,agrobacterium infection time 30 min and termetine solution soaking time 3 min were better.Induction of adventitious buds:MS+4 mg/L 6-BA+50 mg/L Kan+200μmol/L TMT+3.0%sucrose+0.8%agar powder,adventitious bud elongation:MS+4 mg/L 6-BA+50 mg/L Kan+200μmol/L TMT+0.5 mg/L GA_(3)+3.0%sucrose+0.8%agar powder,adventitious bud rooting:1/2MS+50 mg/L Kan+200μmol/L TMT+3.0%sucrose+0.8%agar powder.Conclusion:The genetic transformation plants of BY-1 were obtained and the genetic transformation system of this material was established.
