摘  要:目的:考察核黄素磷酸钠光敏化对人脉络膜黑色素瘤MUM-2B细胞的抑制作用,评价以核黄素磷酸钠为光敏剂光动力学治疗脉络膜黑色素瘤的可行性。方法:首先采用MTT的方法考察核黄素磷酸钠光敏化对MUM-2B细胞的抑制作用,随后利用流式细胞仪从细胞凋亡坏死、线粒体膜电位、周期阻滞3个方面分析核黄素磷酸钠光敏化抑制MUM-2B细胞的机制。结果:单独的核黄素磷酸钠对MUM-2B没有明显的毒性,但是当施以光照辐射,核黄素磷酸钠光敏化能够有效抑制MUM-2B的生长。流式细胞仪分析结果表明核黄素磷酸钠光敏化能够显著提高MUM-2B细胞的凋亡坏死,明显降低细胞线粒体膜电位,同时诱导细胞发生S期和G_(2)的周期阻滞。结论:核黄素磷酸钠光敏化能够有效抑制杀伤人脉络膜黑色素瘤MUM-2B细胞,其具有作为光敏剂进行光动力学治疗脉络膜黑色素瘤的理论可行性。Objective:To investigate the inhibitory effect of RFP-photosensitization on human choroidal melanoma MUM-2B cells,and evaluate the feasibility of RFP as a photosensitizer in the photodynamic treatment of choroidal melanoma.Methods:Firstly,the inhibitory effect of RFP-photosensitization on MUM-2B cells was studied by MTT assay.And then,flow cytometry was used to analyze the effect of RFP-photosensitization on cell apoptosis,mitochondrial membrane potential and cell cycle arrest in order to elucidate related mechanism.Results:When RFP or light irradiation was conducted separately,the survival of MUM-2B cells was hardly affected.However,the synergistic effect of RFP and light irradiation could effectively inhibit the growth of MUM-2B cells.The results of flow cytometry showed that RFP-photosensitization could significantly increase cellular apoptosis and necrosis,obviously reduce the mitochondrial membrane potential,and induce the cell cycle arrest at S and G_(2) phases.Conclusion:RFP-photosensitization exerted significantly anti-tumor effect on MUM-2B cells in vitro,which indicated that RFP has a theoretical feasibility to serve as a photosensitizer in photodynamic treatment of choroidal melanoma.
