摘  要:为研究记忆训练功能是否与小鼠海马区c-FOS蛋白有关,并探究胆碱能受体激动剂和阻断剂与小鼠海马区c-fos基因转录和c-FOS蛋白表达之间是否存在相关性。以小鼠水迷宫学习记忆测试平台为基础,将小鼠随机分为空白组、模型组、逃生组,应用实时荧光定量PCR方法,观察逃生记忆结果与小鼠中枢海马区c-fos基因表达水平变化的相关性。并通过小鼠尾静脉注入毛果芸香碱和阿托品后,测定小鼠海马区c-fos基因转录水平和c-FOS蛋白表达量的变化,探讨对记忆功能有影响的胆碱受体激动剂和抑制剂对记忆的影响是否与c-FOS蛋白表达有关。结果表明:经7d逃生训练试验后小鼠逃生速度极显著大于对照组(P<0.01),且逃生训练组小鼠c-fos基因转录水平极显著高于空白组和模型组(P<0.01),提示小鼠c-fos基因可能与小鼠逃生记忆产生有关。毛果芸香碱可升高小鼠海马区c-fos基因转录水平、阿托品可降低小鼠海马区c-fos基因转录水平,在2.5h达到作用峰值(P<0.01),且具有较好的量效关系;毛果芸香碱可极显著地升高海马区的c-FOS蛋白含量(P<0.01),且此升高作用与捉持和尾静脉注射处理行为无关。从以上结果推测小鼠海马c-FOS蛋白可能与小鼠记忆形成有关。To investigate whether memory training ability is related to c-fos gene in the hippocampus of mouse,the relationship between cholinergic receptor agonists as well as blocker and the transcription of c-fos gene and the expression of c-FOS protein in the hippocampus of mouse was studied.In this study,based on the water maze test platform for learning and memorizing,mice were randomly divided into blank group,model group and escape group.The real-time fluorescence quantitative PCR was applied to estimate the correlation between the results of escape memory and the changes of c-fos gene expression levels in the central hippocampal region of mice.Furthermore,the changes of c-fos gene transcription levels and c-FOS protein expression in the hippocampus of mice were estimated after the injection of pilocarpine and atropine into tail vein.The correlation between the effects of receptor agonists as well as cholinergic blockers on memory function and the expression c-FOS protein was also explored.The results showed that the escape speed of mice after the7descape training test was significantly higher than that of the control group(P<0.01),and the transcription of c-fos gene in the escape training group was significantly higher than that of the blank group and the model group(P<0.01),indicating that the c-fos gene might be related to the production of escape memory in mice.Pilocarpine increased the transcription levels of c-fos gene,while atropine did the opposite with a peak level at 2.5h(P<0.01)in the hippocampus and showed a good dose-effect relationship.Pilocarpine significantly increased the content of c-FOS protein in the hippocampus(P<0.01),and this increase was independent of holding and tail vein injection behavior.From the above results,it could be speculated that c-FOS protein in the hippocampus might be related to the memory formation of mouse.
