摘 要:为探究镉对大鼠睾丸氧化应激和线粒体功能的影响及葛根素的保护效应,将24只雄性SD大鼠随机分为4组,分别为对照组、葛根素组、醋酸镉组、镉与葛根素共处理组,持续5周。试验结束后,制作睾丸病理切片并镜检观察;比色法测定丙二醛(MDA)含量和还原型谷胱甘肽(GSH)活性;使用MTT和Rhodamine 123染料检测线粒体功能;qPCR检测凋亡相关基因表达水平。结果表明:镉导致雄性大鼠睾丸MDA含量极显著升高,GSH活性显著降低(P<0.01),线粒体活力和膜电位降低(P<0.01),凋亡相关基因表达增加(P<0.01),精子数量和活力下降(P<0.01);镉与葛根素共处理组上述损伤明显减轻。这说明氧化应激和线粒体功能障碍是镉致睾丸损伤的关键环节,葛根素可改善镉暴露诱导的睾丸氧化应激、线粒体损伤和细胞凋亡,从而在镉致睾丸损伤中发挥保护效应。To investigate the effects of cadmium on oxidative stress and mitochondrial function in rat testis and the protective effect of puerarin,24 male SD rats were randomly divided into 4 groups,control group,puerarin group,cadmium acetate group and cadmium and puerarin co-treatment group,for 5 weeks.At the end of the test,testicular pathological sections were prepared and examined microscopically;malondialdehyde(MDA)and reduced glutathione(GSH)activity were measured colorimetrically;mitochondrial function was measured using MTT and Rhodamine 123 dye;and apoptosis-related gene expression levels were measured by qPCR.The results showed that cadmium caused a highly significant increase in MDA content(P<0.01),a highly significant decrease in GSH content(P<0.01),a decrease in mitochondrial viability and membrane potential(P<0.01),an increase in the expression of apoptosis-related genes(P<0.01)and a decrease in sperm count and viability(P<0.01)in the testis of male rats;the above damage was significantly reduced in the group co-treated with cadmium and puerarin.It showed that oxidative stress and mitochondrial dysfunction were the critical links of cadmium-induced testicular injury,and that puerarin could improve oxidative stress,mitochondrial damage and apoptosis in testis induced by cadmium exposure,thus exerting a protective effect in cadmium-induced testicular injury.
