摘  要:为探讨长期高脂摄入对大鼠前额叶胆碱能系统的影响及观察有氧运动的干预作用,选择7周龄SD雄性大鼠48只,用高脂饲料喂养建立肥胖大鼠模型,并随机均分为4组,即普通对照组、普通运动组、肥胖安静组及肥胖运动组,运动组大鼠进行8周的有氧跑台运动。试验17周运动结束后宰杀取材,采用比色法测定大鼠血糖(BG)、甘油三酯(TG)、总胆固醇(TC)及前额叶内乙酰胆碱(ACh)含量,采用Western blot方法检测大鼠前额叶乙酰胆碱酯酶(AChE)及胆碱乙酰转移酶(ChAT)的表达情况。结果表明:与普通对照组相比,肥胖组大鼠BG、TG及TC含量极显著升高,前额叶内ACh含量极显著降低,AChE水平极显著降低,ChAT表达无差异;有氧运动后,与肥胖安静组相比,肥胖运动组大鼠BG、TG及TC含量极显著降低,脑前额叶内ACh含量显著上调,AChE表达显著升高,对ChAT表达无影响。这一研究提示长期高脂饮食可导致大鼠血糖、血脂异常,降低肥胖大鼠前额叶胆碱能系统ACh和AChE表达水平;有氧运动能显著改善肥胖大鼠血糖、血脂异常,促进大鼠前额叶ACh和AChE表达。To investigate the effect of high fat diet induced obesity on the cholinergic system in the prefrontal lobe of rats and to observe the regulating effect of aerobic exercise.In the study,obese rats were fed with a high-fat diet for 8 weeks to establish a model.After modeling,obese rats were subjected to aerobic treadmill exercise for 8 weeks.The animals were divided into four groups:control group,exercise group,obesity quietness group and obesity exercise group.Blood glucose(BG),triglyceride(TG),total cholesterol(TC)and the contents of Ach in the prefrontal lobes of rats were determined by colorimetry.The expressions of AChE and ChAT proteins in the prefrontal lobes of rats were detected by Western blot.The results showed that the contents of BG,TG and TC in obese rats fed with high fat diet were significantly increased(P<0.01),the content of Ach in the prefrontal lobe was significantly decreased(P<0.01),the expression of AChE was significantly decreased(P<0.01),and there was no difference in the expression of ChAT.After aerobic exercise,compared with the quiet obesity group(OS),the contents of BG,TG and TC in the obese exercise group(OE)were significantly decreased(P<0.01),the content of Ach in the prefrontal lobe was significantly up-regulated(P<0.05),and the expression of AChE was significantly increased(P<0.05),but the expression of ChAT was not affected.High fat diet can induce obesity in rats,lead to abnormal blood glucose and lipid,and inhibit the levels of related molecules in the cholinergic system in the prefrontal lobe of obese rats.Aerobic exercise can significantly improve the blood glucose and lipid abnormity of obese rats,and promote the expression of acetylcholine and acetylcholinesterase in the prefrontal lobe of rats.
