摘  要:为从基因组水平探究天津猴鸡群体遗传进化过程中的选择信号,补充完善地方鸡种选择信号图谱,发掘天津猴鸡的重要种质特性基因,试验根据纬度分布、遗传关系等条件筛选包括天津猴鸡群体在内的20个地方鸡种及2个引进鸡品种作为研究对象,利用简化基因组测序技术开展基因组测序,分析比较天津猴鸡群体与其他鸡种群体的遗传差异,并对进化过程中受到选择的基因进行基因功能注释。结果显示:在天津猴鸡群体中检测出265869个SNP标记;遗传结构上天津猴鸡群体与狼山鸡等聚为一类,亲缘关系较近;受选择基因主要参与氨基酸生物合成、细胞内信号转导以及神经系统发育等通路或生物学过程;检测到6个与种质特性相关的受选择基因,与神经系统发育、激素调节和抗热应激等生理功能紧密相关。研究结果部分揭示了天津猴鸡的裸颈性状形成和抗热应激特性,为天津猴鸡的保护和种质特性评价提供重要理论依据。This study was aimed to explore selection signals of genetic evolution process of Tianjin-monkey chicken population at genomic level,and improve the selection signal map of indigenous chicken breeds,explore the important germplasm genes of Tianjin-monkey chicken.According to latitude distribution and genetic relationship,twenty indigenous chicken breeds including Tianjin-monkey chicken population and two introduced chicken breeds were selected as the materials.Restriction-site associated sequencing technology was utilized to investigate genomic alterations to analyze and compare genetic difference between Tianjin-monkey chickens population and other chicken breeds.Gene functional enrichment analysis was conducted to investigate the potential biological processes and pathways of the selected genes during evolution.The results showed that 265869 SNP markers were detected in Tianjin-monkey chicken population.Tianjin-monkey chicken population and Langshan chicken were clustered together with closer genetic relationship in genetic structure.The selected genes were mainly involved pathways or biological processes such as amino acid biosynthesis,intracellular signal transduction and nervous system development.Eight selected genes associated with germplasm characteristics were identified,which were closely related to physiological functions such as nervous system development,hormone regulation and anti-heat stress.This study revealed partially naked neck evolution and anti-heat stress character,and would provide an important theoretical basis for bridging genetic resource conservation efforts and the evaluation of germplasm characteristics in Tianjin-monkey chicken population.
