摘  要:目的:探讨维生素D(VDR)基因FokⅠ位点多态性与前列腺增生(BPH)中核因子κB(NF-κB)表达相关性及意义。方法:收集511例手术治疗的BPH患者的血液及组织标本。应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)对上述血液样本进行FokⅠ多态性检测,通过免疫组化方法分析BPH组织标本中NF-κB表达情况。分析VDR不同基因型与NF-κB表达的相关性。结果:NF-κB阳性表达中三种基因型FF、Ff以及ff分布情况为82例(20.7%)、176例(44.3%)、139例(35.0%),NF-κB阴性表达中三种基因型的分布情况:38例(33.3%),49例(43.0%),27例(23.7%),两种表达情况下三种基因型的分布情况差异均无统计学意义(P>0.05),分布符合Hardy-Weinberg定律。FF、Ff以及ff三种基因型中NF-κB阳性率分别为82例(68.3%)、176例(78.2%)、139例(83.7%),阴性率分别38例(31.7%)、49例(21.8%)、27例(16.3%),差异有统计学意义(P<0.05),结论:VDR基因FokⅠ多态性与NF-κB的表达有相关性。Objective To investigate the relationship between single nucleotide polymorphism(SNP) of vitamin D receptor(VDR) gene and expression of NF-κB in BPH.Method The DNA of 511 patients with BPH were abstracted from blood respectively.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was used in determining VDR genotypes.The expression of NF-κB were detected by immunohistochemistry in BPH.The relationship was evaluated between VDR gene polymorphism of FokⅠ alleles and expression of NF-κB.Results Genotype distribution of FF,Ff,ff in group NF-κB positive was 20.7%(82/397),44.3%(176/397),35.0%(139/397)respectively and 33.3%(38/114),43.0%(49/114),23.7%(27/114)in group NF-κB negative.The genotype distribution of two group was in Hardy-Weinberg equilibrium(P>0.05).The ratio of NF-κB positive in FF,Ff and ff genotype was 68.3%(82/120),78.2%(176/225),83.7%(139/166)respectively.The ratio of NF-κB negative in FF,Ff and ff genotype was 31.7%(38/120),21.8%(49/225),16.3%(27/166)respectively.There was significant difference between genotype of VDR and expression of NF-κB(P<0.05).Conclusion VDR FokⅠpolymorphisms is correlated with expression of NF-κB in BPH.
