摘  要:目的 对一例排绦虫节片患者进行驱虫治疗,对虫体进行形态学和分子学鉴定,确定其感染绦虫的种类。方法使用南瓜子-槟榔进行驱虫治疗。对驱出虫体进行形态学观察,采用聚合酶链式法(PCR)扩增虫体线粒体cox1基因片段,对PCR扩增产物经琼脂糖凝胶电泳检测和基因序列测序分析,然后与NCBI数据库中带绦虫cox1基因序列进行比对,同时利用MEGA 6.0软件构建系统进化树。结果 驱出8条长度230~520 cm不等近乎完整的带绦虫虫体,其中5条头节完整,形态学观察疑似亚洲带绦虫或牛带绦虫。PCR扩增cox1基因片段获得大小为1 620 bp的扩增产物,与NCBI数据库中亚洲带绦虫cox1基因(Gen Bank登录号为AB107235.1)同源性为99.58%。进化树分析表明与亚洲带绦虫的亲缘关系最近。结论 该病例属于亚洲带绦虫感染。Objective To provide treatment for a patient infected with tapeworms and identify the species of the tapeworms by morphological and molecular methods. Methods Areca and pumpkin seeds were used for deworming. Morphological observation,PCR amplification and sequencing of cytochrome coxidase 1(cox1) gene were performed for identifying the discharged worms.BLAST alignment analysis on the obtained target sequences was carried out with the published cox1 sequences of T. asiatica.The genetic distance of obtained target sequences was calculated by MEGA 6.0, and phylogenetic tree was constructed by Neighbor-Joining. Results Eight almost complete tapeworms(from 230 to 520 cm in length) were obtained after treatment. Of them, five had complete head section. The discharged worms were suspected to be Taenia saginata or T. asiatica by morphological observation. PCR amplification of cox1 resulted in a band of 1 620bp, which was 99.58% homologous to cox1 of T. asiatica(Gen Bank accession number: AB107235.1). Phylogenetic tree analysis showed that it had the closest genetic relationship with T. asiatica. Conclusion According to the clinical feature and sequencing results, this case is confirmed to be a local infection of T. asiatica.
