摘 要:八仙花(Hydrangea macrophylla)花序硕大,花色多样,变种多,观赏价值高,国内外广泛栽培。本研究开展了八仙花的一步成苗离体培养及组织器官激素富集研究。结果表明:MS+1.0 mg·L^(-1)6-BA+0.2mg·L^(-1)NAA为八仙花茎段一步成苗诱导最佳配方,首次建立了八仙花一步成苗离体再生技术体系。八仙花受试组织器官6-BA富集水平依次为叶片>茎段>叶柄>茎尖,NAA富集水平依次为叶片>叶柄>茎尖>茎段。本实验在同一培养基条件下,受试材料活力高,无需更换培养基,由外植体一次性发育为完整植株,操作简单,成本低廉,适宜规模化生产。Hydrangea macrophylla with huge inflorescences and various colors has high ornamental value and is widely cultivated all over the world.This study carried out the one-step seedling culture of hydrangea in vitro and the characteristics of hormone enrichment in different tissue organs.The results showed that MS+1.0 mg·L^(-1) 6-BA+0.2 mg·L^(-1) NAA was an excellent one-step seedling formation induction formula,with stems of hydrangea as explants.This research established a technical system for one-step regeneration of hydrangea seedlings in vitro for the first time.The concentration of 6-BA in the tested tissues and organs of hydrangea was leaf>stem>petiole>stem tip,and the NAA enrichment level was leaf>petiole>stem tip>stem.Under the same culture medium condition,the test material had high vitality,no need to change the culture medium,and the explant grew into a complete plant at one step.The operation is convenient and it is suitable for large-scale production.
