摘 要:糖酸组分与含量是影响石榴果实品质的重要指标,研究其代谢机理能为石榴品质改良提供理论依据。本研究用Illumina高通量测序技术比较分析了石榴‘泰山红’和‘牡丹’成熟期的转录组数据,对糖酸代谢相关基因进行了鉴定和表达分析。结果表明,共获得51670个差异表达基因,其中显著差异表达基因2409个,不显著差异表达基因40101个。在GO数据库中,7502个差异基因被注释到3个类别46个亚类中,主要参与代谢过程和细胞过程。在KEGG代谢通路中,257个差异基因参与了19个代谢通路,代谢途径和次生代谢物生物合成途径得到显著富集。通过生物信息学分析,挖掘出参与石榴果实糖酸代谢的关键基因17个。qRT-PCR分析表明,2个石榴品种中TK基因的表达量最高,存在品种差异性。‘泰山红’中FRK、SS、BGLU、UXS、FBA和ADH的表达量高于‘牡丹’。石榴果实中,葡萄糖和果糖是主要的糖组分,酸以柠檬酸和苹果酸为主。The composition and content of sugar and acid are important indicators that affect the quality of pomegranate(Punica granatum) fruit,and studying its metabolic mechanism can provide a theoretical basis for the improvement of pomegranate quality.In this study,Illumina high-throughput sequencing technology was used to compare and analyze the transcriptome data of ‘Taishanhong’ and ‘Mudan’ at the mature stage.The expression of genes related to sugar and acid metabolism was identified and analyzed.The results showed that a total of 51 670 differentially expressed genes were obtained,of which 2 409 were significantly differentially expressed genes and 40 101 were not significantly differentially expressed genes.In the GO database,7 502 differentially expressed genes were annotated into 3 categories and 46 subcategories,mainly involved in metabolic and cellular processes.In the KEGG metabolic pathway,257 differential genes were involved in 19 metabolic pathways,and the metabolic pathways and secondary metabolite biosynthetic pathways were significantly enriched.Through bioinformatics analysis,17 key genes involved in the metabolism of sugar and acid in pomegranate fruit were identified.The q RT-PCR analysis showed that the TK gene expression level was the highest in two cultivars,and existed cultivar differences.The expression levels of FRK,SS,BGLU,UXS,FBA and ADH in ‘Taishanhong’ were higher than that in ‘Mudan’.In pomegranate fruit,glucose and fructose are the main sugar components,and the acids are mainly citric acid and malic acid.
